Objective:This essay aims to help eliminate the discrepancies in the classification of Liriope spicata Lour. var. prolifera Y. T. Ma, and lay a foundation for the naming of non-scientifically-named plants.Methods:1Collect Liriope spicata Lour.var.prolifera Y.T.Ma, no scientific name plants and some other plants which is more likely to be the parent or the original variant plants, cultivat them in pots, and extract genomic DNA from their fresh leaves.2Find the optimal reaction system and reaction conditions for the extraction of ISSR through orthogonal design method.3Randomly select52ISSR primers from the ISSR primer sequence table, and then from which filter10highly polymorphic primers.4Analyze all the selected plants of extracts through the method of ISSR.5Convert the result of ISSR into0and1data matrix, and calculate it with software spss16.0so as to get similarity coefficients of any two selected plants, and constract of the study.6Speculate the research conclusion according to the similarity coefficients, cluster map and morphological classification results under the guidance of the taxonomy, genetics underlying theories。Results:Establish a stable reaction,strip clear optimal ISSR reaction system and amplification procedure through the orthogonal experiment. Screen out annealing temperature of10primers;10primers were amplified from141bands, including140polymorphic, ratio of which was99.29%.The primer amplification bands were11-17; The cluster diagram shows the plants of the same name but from different producing areas were priorly clustered in addition to Liriope muscari (Decne.) Baily from Quanzhou city and Liriope muscari (Decne.) Baily from Lushan Mountain.The similarity coefficient also shows a correlation of each of the two plants of the same name of them, more than the similarity coefficient of the plants with same name. The Liriope muscari (Decne.) Baily from Lushan Mountain priorily clustered with Liriope. spicata(Thunb.)Lour, in the dendrogram,which was followed with the same name but from different origins of the Liriope muscari (Decne.) Baily from Quanzhou city.The similarity coefficient of Liriope muscari (Decne.)Baily from Lushan Mountain No.5and No.14, No.15Liriope. spicata (Thunb.) Lour.is0.3375which is almost equal to the average number of similar coefficient which was0.338between No.5and No.6. The similarity coefficient of Liriope spicata Lour.var.prolifera Y. T. Ma and non-scientifically-named plants were0.373and0.415which were more than Liriope spicata Lour.var. prolifera Y. T. Ma and other selected plants similarity coefficient;The similarity coefficient of Liriope spicata Lour. var. prolifera Y. T. Ma and Liriope.spicata(Thunb.) Lour, was0.279and0.348, the average number of similar coefficient was0.313. similarity coefficient of Liriope musacari (Decne.) Bailey from Lushan Mountain and Liriope spicata Lour. var. prolifera Y. T. Ma was0.319, and that genetic distance of Liriope spicata Lour.var. prolifera Y. T. Ma and Liriope spicata Lour, basically equal to genetic distance of Liriope spicata Lour. var. prolifera Y. T. Ma and Liriope muscari (Decne.) Baily from Lushan Mountain. The similarity coefficient of Liriope spicata Lour. var. prolifera Y. T. Ma and Liriope spicata Lour, or Liriope muscari (Decne.) Baily from Lushan Mountain was smaller than that of Liriope spicata Lour.var. prolifera Y. T. Ma with the rest of the selected plants (i.e., Liriope graminifolia (L.) Baker, Liriope platyphyla Wang et Tang or Ophiopogon japonicus (L.f) Ker Gawl).Conclusion:1Although the similarity coefficients of Liriope spicata Lour.var. prolifera Y. T. Ma and non-scientifically-named plant and the rest of the plants but since these two kinds of plants are all flowers, they cannot be treated as the other party or variations of the variants.2The similarity coefficient of Liriope spicata Lour.var. prolifera Y. T. Ma and Liriope. spicata(Thunb.) Lour.ov the similarity coefficient of Liriope musacari(Decne.) Bailey from Lushan Mountain are second to that of non-scientificlly-named plants in size, taking into account of the features that it has flowers we can infer that Liriope spicata Lour.var. prolifera Y. T. Ma is the hybrid of Liriope musacari (Decne.) Bailey from Lushan Mountain and Liriope.spicata(Thunb.) Lour.3The non-scientifically-named plants were the variant of Liriope. Spicata (Thunb.) Lour.for their similarity coefficients with Liriope. spicata(Thunb.) Lour.are second to that of Liriope spicata Lour.var. prolifera Y. T. Ma.4The genetic distance of Liriope. muscari (Decne.) Baily from Quanzhou city is the variant of Liriope. muscari (Decne.) Baily from Lushan Mountain, the reason is:Liriope. muscari (Decne.) Baily from Lushan Mountain did not cluster priorily with the Liriope. muscari (Decne.) Baily from Quanzhou city.The genetic distance of Liriope. muscari (Decne.) Baily from Quanzhou city and Liriope. muscari (Decne.) Baily from Lushan Mountain is shortest, which is almost equal to the genetic distance between Liriope. muscari (Decne.) Baily from Lushan Mountain and Liriope spicata(Thunb.)Lour. Liriope. muscari (Decne.) Baily from Lushan Mountain is wild,while Liriope. muscari (Decne.) Baily from Quanzhou city is cultivated; Liriope. muscari (Decne.) Baily from Lushan Mountain leaves are leathery, but Liriope. muscari (Decne.) Baily from Quanzhou city leaves was not leathery,and leaves are leathery is one of the characteristics of Liriope. muscari (Decne.) Baily.
