Objective To study the mechanism of electro acupuncture against apoptosis and gene control of acute focal brain ischemia in rat. Methods SD rats were divided into four groups: normal group, model group, sham operative group, electroacupunction group. Block the rats' middle cerebral artery with line peg method induce focal brain ischemia in rats. "Shuigou'V'Neiguan", "Baihui" were electroacupunctured. By the experimental technology of neuropathology, neurobiochemistry, molecular biology, the nerve missing sign, pathological changes of neurons in ischemic cortex, sum of ischemic apoptosis neuron is ischemic cortex, the protein expression of bc1-2 bax. P53 . caspess-3. Fas in is chemic cortex, and the effects on above item of electro acupuncture were observed. Results (l)Electro acupuncture could significantly improve the nerve missing sign of rats with focal brain ischemia (P<0.05), release the number of apoptosis neurons in ischemic cortex (P<0.01) and improve pathological changes of neurons in ischemic cortex. (2)Electro acupuncture could significantly promote the protein expression of bcl-2 which in model rats has been significantly low(P<0.05), and decrease the protein expression of bax which in model rats has been significa ntly high (P<0.05). (3)Electroacupuncture could significantly reduce the protein expression of P53 which in model rats have been significantly higher than normal levels (P<0.05).(4)Electroacupuncture could significantly reduce the protein expression of caspase-3 which in model rats have been significantly higher thn nomal levels (P<0.01). (5)Electroacupuncture could significantly decrease the protein expression of Fas which in model rats have been significantly higher than normal levels (P<0.05). Conclusion Genes bcl-2. bax. P53> caspase-3. Fas participate in apoptosis afterischemic brain injury Electroacupuncture has the effect against ischemic brain injury of focal brain ischemia. This effect of electroacupuncture could be carried out by following ways: Promote the protein expression of bc1-2 which is anti-apoptosis gene. Reduce the protein expression of bax which is proapoptosis gene. Decrease the protein expression of P53 which is proapoptosis gene. Decrease the protein expression of caspase-3 which is proapoptosis gene. Decrease the protein expression of Fas which is proapoptosis gene. Furthermore electroacupuncture could offectivly apoptosis which is led by ischemicbrain injury after focal brain ischemia...
