Objective:To research the methods of external therapy for hyperplasia of mammary gland(HMG) with Traditional Chinese Medicine,and obserbv the mechanism of action of Runingshuang in curing HMG.Methods:Combining the Traditional Chinese Medicine theory to the modern pharmaceutic technology,with the principle of regulating qi and promoting blood flow,removing obstruction of channels and resolving mass,reducing swelling and arresting pain,Runingshuang was produced precisely.Permeable rate of Runingshuang was measured with by liquid chromatography on separating mouse skin.In the research , animal pattern of HMG was made by estrone and progesterone injection.Rats were randomly divided into 7 groups: the comparative group,the animal pattern group,the tamoxifen group,the Sanjieruipi Tiegao group,groups of high,middle and low dosage of Runingshuang.These groups were treated with different drugs.Observing the shape of breast,microcirculation,blood rheology,sexual hormones,pathological changes of breast tissue,and the expression of VEGF,bFGF,PCNA,Bcl-2,bax,as well as tissue apoptosis.Result:As the basis of Runingshuang,the penetrating rate in 24h of eugenol was 17.29%,which indicated the good penetrating absorbing effects of Runingshuang. As was shown in the experiment,all of the high,middle and low dosage of Runingshuang could restrain intumescence of rats'breasts with HMG(P<0.01),improve breast microcirculation perfusion.Effect of both high and middle dosage was better than Sanjieruipi Tiegao(P<0.05),and had no difference with tamoxifen(P>0.05).Runingshuang could reduce blood viscosity(P<0.05 or 0.01) , which was in advance of tamoxifen(P<0.01),and reduce the content of serum E2 and PRL,and increase the the content of serum P(P<0.01),and lighten breast hyperplasia(P<0.01).As was shown in theimmunohistochemical research , Runingshuang(including high , middle and low dosage)could reduce the expression of tissue ER,PR and VEGF(P<0.01),reduce the expression of PCNA (P<0.05), with no difference among the three groups(P>0.05).Runingshuang of high and middle dosage could increase the expression of bax(P<0.05),and reduce the expression of Bcl-2(P<0.01).Conclusion:It is an effective method to treat HMG with transdermal therapeuty of Runingshuang. The curing mechanism of Runingshuang is to improve breast microcirculation,increasing blood perfusion, meliorate blood hypercoagulability,regulate incretion,reduce the hyperplasia of mammary tissue.In addition,Runingshuang can cure mammary hyperplasia by decreasing the expression of VEGF and bFGF.It can also accelerate apoptosis by regulating the expression of PCNA and Bcl-2.This study has enriched the external therapy and experimental methods of HMG, and laid foundation for drug exploiture.
