Objective: To explore the influence of smoking on the function and expression of human bone tissue osteopontin(OPN) .Methods : Each 10 volunteers of smoking and non-smoking healthy adult tibia fracture were randomly divided into two groups: smoking group and non-smoking group. The patients in the smoking group were living with daily smoking above 20 cigarettes and had been smoking at least 10 years. The patients in non-smoking group were living with daily smoking no more than 1 cigarettes or no smoking. Each of them were took abour 1g cancellous bone in proximal tibia metaphysis and the total protein and the total RNA were extracted from them. OPN mRNA expression were measured by RT-PCR, OPN expression were measured by Western blotting and OPN phosphorylation degree weremeasured by immuno-precipitation.Results : The OPN/ beta -actin ratios of band optical density values in smoking group RT-PCR are higher than the OPN/ beta -actin ratios of band optical density values in non-smoking group RT-PCR and the differences is significant (P < 0.05).
