Objective: To investigate the effect of bifidobacteria andlactobacilli on mice dextran sulphate sodium (DSS)-induced acuteulcerative colitis. To research the potential mechanism for probioticstherapy of dextran sulphate sodium (DSS)-induced acute ulcerative colitis,and to observe expression of IL 10, GR and HSP70 in the colon mucosa.Methods:(1)To select effective probiotics strains to treat ulcerative colitis byinjiciatur enema. Mouse ulcerative colitis was induced by dextransulphate sodium(DSS). Balb/c mice were randomly divided intofollowing group: normal control group, model group, negative controlgroup, positive control group, DSS add bifidobacteria CMS-H004 straininjiciatur enema group, DSS add bifidobacteria CH22 strain injiciaturenema group, DSS add lactobacilli CMS-H002 strain injiciatur enemagroup and DSS add lactobacilli f3-5 strain injiciatur enema group. Toobserve each mouse's general state of health on experiment procese. Allmice were sacrificed in 9th day, to obtaine colon tissue specimens and tomeasure length of integrity colon. Theexpression of interleukin -10 (IL-10), GR and HSP70 protein weredetected among cells of colon tissue with immunohistochemical stain andWestem-blotting, and using RT-PCR to detect to mRNA expression ofIL10, GR and HSP70.Result:(1)Mice's colon inflammation degree of DSS add bifidobacteriainjiciatur enema groups were aggravated than that of model group, andmice's colon inflammation degree of DSS add lactobacilli injiciaturenema groups were mitigated than that of model group(2)Mice's colon inflammation degree of DSS add bifidobacteriainjiciatur enema groups were aggravated than that of model group, butcolon inflammation degree of mice merely gave an enema ofbifidobacteria weren't aggravated than those of normal control.Expressions of IL-10 and HSP70 of DSS add bifidobacteria injiciaturenema groups were reduced compared with that of model group, andexpressions of IL-10 and HSP70 of mice merely gave an enema ofbifidobacteria weren't reduced than those of normal control.(3) Lactobacilli injiciatur enema cause increased expression ofHSP70 and IL10 on colon mucosa of mice dextran sulphate sodium (DSS)-induced acute ulcerative colitis.Conclusion:(1)Some strains of bifidobacteria aggravate mice's coloninflammation, and reduce expressions of IL-10 and HSP70 when mucosalbarrier of colon were destroyed.(2)Some strains of bifidobacteria possible are opportunistic pathogenof ulcerative colitis.(3) Lactobacilli was also associated with a reduction in mucosalinflammatory activity and lesion scores.
