Object:To observe the effect of SHENMAI injection on Focal Cerebral Ischemia Reperfusion Injury rats, this study was designed to observe ischemic penumbra angionegesis,neural proliferation of mice, in order to SHENMAI injection provide a theoretical basis for.the clinical wide range of application.Methods:All of male and female SD rats were randomly divided into blank group, MCAO/R model group, PNS (total saponins of Panax notognseng) control group, Shenmai injection therapy group. According to different drawing materials time there was 3 days,7 days, and 14 days time point after cerebral ischemia in each group,with 6 cases in each time point.Production suture method in rats with focal cerebral ischemia-reperfusion model was successful in each group were randomly taken 2, TTC staining to determine the results of modeling. The blank group were not given any intervention, and model group were given 0.9% NS injections,3,7,14 days after treatment rats were killed. Perfusion-fixed brain tissue, HE staining of pathological changes and immunohistochemical observation of VEGF Brdu CD34 expression, transmission electron microscopy observation of ultrastructural changes. RT-PCR test, observed treatment rats were killed after 3,7,14 days, take fresh brain tissue.observed VEGFmRNA expression. SHENMAI group and PNS group significantly increased positive expression rate of VEGF\Brdu\ CD34 and VEGFmRNA; but three days time period, the positive expression rate about VEGF\Brdu\CD34 and VEGFmRNA in PNS group are better than SHENMAI group; 7 days time period, between SHENMAI group and PNS group, the positive expression rate of VEGF\CD34 and VEGFmRNA are same hardly; 14-day time period, the positive expression rate of Brdu in SHENMAI group better than PNS group, and positive expression rate of VEGF\CD34 and VEGFmRNA in SHENMAI group compared with PNS group have no significant. Conclusions:①Shen Mai injection can significantly promote the expression of vascular endothelial growth factor (VEGF) and VEGFmRNA in the ischemia penumbra of the brain in the rats, but in the acute phase (3 days) the effects is weaker than PNS. Its main effects in recovery.②Shen Mai injection can significantly increased microvascular density in the ischemia penumbra of the brain in the rats, but in the acute phase (3 days) the effects is weaker than PNS.③Shen Mai injection can significantly promote the Brdu number of positive cells in the ischemia penumbra of the brain in the rats, half of acute (7 days) and recovery start time (14 days) more apparent.④Shen Mai injection can protect cerebral ischemia and reperfusion in rat neuronal ultrastructure of mitochondria and rough endoplasmic reticulum.⑤Shenmai injection can significantly promote cerebral ischemia asymmetric sensorimotor disorder recovery, and neurological score improved with treatment "time of Shen Mai injection correlated.
