Objective: Ulcerative colitis is a kind of nonspecific inflammatorybowel diseases (IBD) .The etiology and mechanism of IBD are not clearcompletely and the curative effect is not satisfied, therefore it isdifficult to be cured in clinic. There is a long history and much experiencein treatment of ulcerative colitis with Traditional ChineseMedicine (TCM), it is worthy to be studied for us. Although there are a lotof clinical and experimental studies about immune function disturbanceand inflammatory reaction in treatment of ulcerative colitis with TCM , thecurative method is individual experience, and therapy shorts ofcomparative study. Advantages of TCM in prevent and treatment ofulcerative colitis are reducing recurrence and enhancing long curativeeffect. according to theory of TCM and clinical prescriptions, the basiccurative method belongs to category of YiQi, HuoXun, JianPi, BuShen. In thispaper comparative studies for curative method ofYiQi, HuoXun, JianPi, BuShen in prevent and treatment of dextran sulfatesodium-induced Mice ulcerative colitis Model were done, and the curativeeffect and mechanism were observed from cytokine, expression of mRNA andprotein. It is important for clinical and academy significance to explorecurative mechanism of chronic inflammation on providing experimentalevidence of TCM in prevent and treatment of ulcerative colitis.Methods: (l)The pathlolgical model of acute colitis was induced by feeding with 3%DSS for 7 days in mice. 100 Balb/c mice were randomly divided intosevengroups, namelynormalcontrolgroup, thepathologicalgroup, solfasa lazine(SASP)group, BaoYuanTang(BYT)group, TaoHongSiWuTang(THSWT)group, S hengLinBaiShuSan(SLBBS)group,JinGuiShenYiWan(JGSYW) group. There were 10 mice in normal conarol group and 15 mice in other groups. the six mice model groups of UC were treated by distilled water, SASP, BYT, THSWT, SLBBS, JGSYW respectively. they were fed with 3%DSSafter 3 days and sequentially treated by drugs for 7 days meanwhile. After those treatments, the changes of symptoms and weight , disease activity index, colonic histologic score , the level of IFN-γ, IL-4, IL-10 in serum, spleen,colon tissue, the expression of TNF-α , iNOS in the colon tissue were observed and detected.(2) chronic colitis was induced by feeding with 3%DSS for 7 days and then drinking water in the following 14 days in mice. 100 Balb/c mice were randomly divided into seven groups when chronic colitis was induced successful, they were given the same treatment which described above. The expression of NF- κ BP65,COX-2,TGF-β, EGF were examined by immunohistochemistry,the NF-κ BP65 mRNA,C0X-2mRNA,TGF-β mRNA,EGFmRNA by hybridization in situ.Results:1. Following results came from experiment of acute colitis model in mice . (l)the disease active index and colonic histologic score in pathlolgic group were significantly higher than those in normal group (P<0. 05), the disease active index, colonic histologic score in SASP group, BYT group, THSWT group, SLBBS group, JGSYW group were lower than those in pathlolgic group(P<0.05). (2)the expression of TNF-α,iNOS and level of IFN-γ in colon tissue in pathlolgic group were obviously higher than those in normal group(P<0.05), the expression of TNF-α , iNOS and level of IFN-γ in colon tissue in SASP group, BYT group, THSWT group,SLBBS group,JGSYW group were lower than those in pathlolgic group (P<0.05);But the changes of level of IL-10 was opposite.2. Following results were from experiment of chronic colitis model in mice. (1) colonic histologic score in pathlolgic group was significantly higher than those in normal group(P<0. 05), colonic histologic score in SASP group, THSWT group, JGSYW group were lower than those in pathlolgic group(P<0. 05).
