Objectives: Based on the summary and analysis of the current documents, the study explores roles of phlegm and stasis in the formation of Hyperlipidemia. By analyzing the influence of the Huatan Huoxue Fang (HHF) on experimental Hyperlipidemia in rats and on bovine aorta endothelial cells (BAEC), the author goes further to sort out the effect of the HHF in the treatment of Hyperlipidemia and its working mechanism.Methods: Theoretical Inquiry: The methods of documents collection, summary, and analysis are adopted. Experimental Inquiry: A. The influence on the experimental Hyperlipidemia in rats, the rats are sub-divided randomly into the normal control group, the model group, Fenobrite group, and finally, big, middle, and small groups according to HHT. 1.Use enzyme methods and immunization photoextinction individually to detect the levels of total cholesterol (TC) , triglyceride (TG), low density lipoprotein cholesterol (LDL-c), high density lipoprotein cholesterol (HDL-c), HDL2-C apolipoprotein A (ApoAI)and apolipoprotein B (ApoB100)in blood, use 2% -16% non-denaturing poplyacrylamide gradient gel electrophoresis to detect the plasma level of small dense low-density lipoprotein (sdLDL). 2. The observation of the improve of hemorheology by testing whole blood viscosity, plasma viscosity and red-cell specific volume; 3. Use enzyme methods to detect superoxide dismutase (SOD), malondialdehyde (MDA) and nitric oxide (NO) content. 4. Use optical microscopes to monitor the morphologic change of liver. 5. Use immunohistochemistry assay to detect the expressed level of low density lipoprotein receptor (LDL-R) in liver.6. Use RT-PCR to get the levels of low density lipoprotein receptor mRNA (LDL-RmRNA), peroxisome proliferators actovated receptor alpha mRNA (PPARamRNA) and acyl-CoA oxidase mRNA (ACOmRNA) in liver. B. The influence on BAEC, the serum containing HHF of different concentration ( 5% 10% 20%) add to the 3-5 era BAEC, 10 hours after applying Western Blot, sort out the influence of HHF on BAEC endothelial nitric oxide synthase (eNOS) based on the level of protein.Results: In theoretics study, the literatures point out that disorder of diet and seven emotional factors and organs of ZangFu and phlegm and stasis are its major etiological factors. No matter how human body is influenced by internal and external factors, hyperlipidemia presents itself as the formation of phlegm and stasis. The method of Huatan Huoxue as the treatment for Hyperlipidemia conforms to the fundamental theory of Chinese traditional medicine. Experimental Result: HHF has the following effects: 1. It obviously reduces the levels of TC, TG, LDL-c, sdLDL, ApoB100 in rats' blood, showing obvious difference from the model group( P<0.05 or P<0.01 ) , it increases the levels of HDL-c, HDL2-C, and ApoAI in rats' serum, showing obvious difference from the model group (P<0.05 or P<0.01 ) . 2. It obviously reduces the levels of whole blood viscosity (low high ) plasma viscosity and red-cell specific volume, showing obvious difference from the model group (P<0.01 ) . 3. It obviously increases the level of NO and SOD, and decreases thelevel of MDA in rats' serum (P<0.01). 4. It obviously increase the presence of LDLR in rats' liver of model group, showing obvious difference from the model group (P<0.05). 5. It obviously increases the presence of LDL-RmRNA, PPARamRNA and ACOmRNA in rats' liver. 6. The pathology shows that, in the comparison group of the rats, the morphological structures of the livers are normal. Livers lobule is regular, no lipid drop is found, in the modal group, many abnormal symptoms are visible: liver cells are swollen, lipid drop of various sizes are found on the liver, in the treatment group, liver has mild and less lipid drops. 7. The serum of HHF obviously can increase the level of eNOS protein on the internal cells of the main artery.Conclusion: 1.
