Objective: In the article of study,the reasearcher summed up and inherited the treatment experiences who professor Ren jixue,a famous Chinese physician on treating acute ICH by TCM. Ultimately, to studying the treatmental mechanism of xingnao jianshen capsule(professor Ren jixue's experiential prescription on treating acute ICH) to cure ICH on molecule level.The acute ICH is a mostly killer to human health, the prognosis is not ideal. Therefore, to seek a sort of TCM that has abroad application foreground to cure the acute ICH is imperative under the situation. Traditional Chinese medicines was more effective in curing ICH, it should be studied further.Professor Ren jixue is a famous doctor of TCM on acute disease of TCM, especially, he is good at treating acute brain disease of TCM. The study of experiment and clinical on acute ICH by treatment of Traditional Chinese medicines,that prosser Ren jixue has preside an item on "eight-five" period, has won the thrith grade prize of the state science-technique advancement prize. The experimental researche has approved that xingnao jianhsen capsule(professor Ren jixue's experiential prescription on treating acute ICH) has an obvious effect on protecting brain ptasmalemma and blood-brain barrier; moreover, it also can improve the activity of SOD into brain tissue; rectify the metabolizing turbulence of free radical and improve ion distribution; can resist cerebral edema. Although the experimental research on acute ICH by xingnao jianshen capsule has acquired some success and obtained significant clinical effect, but the complete effect mechanism of xingnao jianshen capsule on acute ICH should be researched deeply.In recent,the study of approved that the value of serum and GSF for neurotissue protein S100B(S100B protein,mostly lie in gliai cell) and Neuron specific enolase(NSE, mostly lie in nerve cell) assue dynamic change. These studies mostly centralized on the relation between the hemorrhage volume, nerve function defect, prognosis and the change of NSE and S100B. the study approved their value of serum and GSF were positive correlation with the degree of brain damage, the two sensitivity neurobiochemical markers have been used to estimating the degree of brain damage and prognosis. Besides the change of the level of serum and GSF NSE and S100B relate to leakage after brain damage, whether it also relate to gene controlling level should be studied further too. Furthermore, the study,which regarding NSE and S100B as index to evaluate whether xingnao jianshen capsule has therapeutic action, has not beenexperimentized at all times, the researcher of article mainly studied academic experiences that professor Ren Jixue treated ICH and discussed his distinctive theoretical system, at base of theorey, the researcher mainly regard the neurobiochemical markers(NSE and S100B) and experimental wistar rats as research objects by some modem western medicine techniques including ELISA, ABC Immunohisto-chemical assay, RT-PCR technique,moreover, by observing the change of expression mRNA and protein level for the sensitivity neurobiochemical markers(NSE and S100B) in experimental acute intra-cerebral hemorrhage of rats of different group at different points of time; Ultimately, to studying the treatmental mechanism of xingnao jianshen capsule to cure ICH on molecule level. Mainly purpose of the researcher lied in inheriting systematically the theories of traditional Chinese medicines and taking it as guiding principles of clinical and scientific researches. Secondly, it can widen academic insights and enhancing clinical curing effects by inheriting precious experiences of the famous doctor of TCM so as to go step further to improve curative effect of the knotty disease. On the other hand, to explore the treatmental mechanism of xingnao jianshen capsule on molecule level on ICH.Method 72 healthy wistar rats were divided into three groups at random. A group(ICH+xingnao jianshen capsule group)(n=24): xingnao jianshen capsule(30mg/100g) were given by oral and injected into abdominal cavity after operation two times a day. B group(ICH+NS group)(n=24),0.9% Nacl(2ml) were injected into abdominal cavity after operation two times a day. C group(sham operation group)(n=24): only food were generaly feeded after operation. Rat model of acute intracerebral hemorrhage were made by injection of self-body blood into the right caudatus-putamen nucleus of rats. Sham operation group do not inject blood into the cerebrums. The lesioned degree of neuro logical function was assessed by zea-longa score. The rats were allowed to survive for 1, 3, 7, 14days. Six rats were killed at above each different points of time. The following examinations were carried out at 1,3, 7, 14d after operation.(1), The lesioned degree of neuro logical function was assessed by zea-longa score.(2), Their seril blood S-100B and NSE were measured by ELISA.(3),By using the RT-PCR technique, the expression of mRNA for S-100B and NSE in the region of perihematoma at different points of time after ICH was tested.(4), ABCImmunohisto-chemical assay was used to investigate the changes of the expression forS-100B and NSE at the protein level, immune positive cells were stained brown-yellowcolour in the cytoplasm. (5),The number of NSE and S-100B positive cells were detected withLSCM under 400×object lens continue 10 eyeshots, taking count of the quantity of positivecells in each high times eyeshot and calcute average at the same time.Result: The experiment indicated: (1),there were no significant differences in theneurological deficit score between A group and B group after operation (p>0.05), but Cgroup was significant decreaseer on the NDS than A group and B group after operation. Atevery time points, there were significant differences in the NDS between three group afteroperation (p<0.01); Moreover, in every group the NDS were no differences afteroperation.(p>0.05); but A group was significant decreaseer on the NDS than B groupfrom3d, there were significant differences between A group and B group(p<0.05); therewere more significant differences from 7d to 14d between A group and B group(p<0.01).(2),The values of serum S-100B levels of B group were significantly higher than those ratsof C group at different points times from 1d~7d (p<0.05). A peak of serum S-100B emerged at3d after ICH. there were markedly difference between B group and C group (p<0.01); After7d, it decreased gradually, but it maintained higher level at 14d. there were significantlydifferences between B group and C group(p<0.05); After treatment, the level of serumS-100B in the rats treated with xingnao jianshen capsule dropped than B group at differentpoints times, but there were no markedly difference between them on level of serum S-100Bat 1d (p>0.05).; there were markedly more difference between A group and C group from 1dto 3d (p<0.01); there were markedly difference between A group and B group from 3d(p<0.05); there were markedly difference between A group and C group from 7d (p<0.05);there were markedly difference between A group and B group from 7d(p<0.01), there werenot significantly differences between three groups on level of serum S-100B at 14d (p>0.05).(3),The expression of mRNA of C group were not significant at different points times. Theexpression of mRNA of B group markedly increased at 1d. The peak of the expression ofS100B mRNA emerged at 7d. After 7d, it decreased gradually, At 14d, the level of theexpressions of S100B mRNA closed to sham operation group at corresponding points time; After treatment, the expression of mRNA of S-100B in the rats treated with xingnao jianshencapsule dropped markedly than B group at 1d, meanwhile, it was lower than C group atcorresponding points time; After 1d, it increased gradually, at 3d, it closed to C group atcorresponding points time; The peak of the expression of S100B mRNA emerged at 7d.furthermore, it was higher than C group at this time, The expression of mRNA of S-100B inthe rats treated with xingnao jianshen capsule were lower than B group from 1d to 7d. Theexpression of mRNA of S-100B between three group were not significant at 14d. (4),①Thepositive cells dyed darkly and the quantities of positive cells of B group increased, there weresignificantly differences between B group and C group (P<0.05); At 3d, the volume of cellsincreased, immunoreaction gradually increased, at meantime, the quantities of positive cellsof B group increased too, there were significantly differences between B group and C group(P<0.01); A peak of immunoreaction emerged at 7d after ICH, the gliai cells swelledmarkedly and protuberance largend significantly, assumed rotundity or anomaly form; atmeantime, the quantities of positive cells of B group increased significantly too, There weremore significantly differences between B group and C group (P<0.01); The immunoreactionweakend(stain becomed light) from 14d, there were more significantly differences betweenB group and C group; Although the quantities of positive cells of B group high little than Cgroup at 14d, there were more not significantly differences between B group and C group(p>0.05);②The cells form of A group changed no significantly, the quantities ofprotuberance more less; the stain was more dark than C group at different time pointsfrom 1d to 7d, but the stain was more lighter than B group at different time points fromld to7d, There were not significantly differences between three groups at 14d; The quantities ofpositive S-100B cells C group were significantly lower than those rats of B group at differentpoints times from 1d to 7d, there were significantly differences between A group and Bgroup(p<0.01); one of three groups, the quantities of positive S-100B cells decreased mostsignificantly at 1d; There were not significantly differences between three groups (P>0.05);③The quantities of positive S-100B cells of A group more lower than C group, There weresignificantly differences between A group and C group(p<0.01); The quantities of positiveS-100B cells of A group increased gradually at 3d, but there were not significantly differencesbetween A group and C group (P>0.05); A peak of the quantities of positive cells emerged at 7d after ICH, there were significantly differences between A group and C group (P<0.05);The quantities of positive S-100B cells of A group decreaed markedly at 14d, there were notsignificantly differences between A group and C group (P>0.05); (5),①The values ofserum NSE levels of B group were significantly higher than those rats of C group at differentpoints times at 1d(p<0.05). A peak of serum S-100B emerged at 3d after ICH. there weremarkedly difference between B group and C group(p<0.01); After 3d, it decreased gradually,but it maintained higher level at 7d. there were significantly differences between B group andC group (p<0.05); at 14d, the values of serum NSE of B group decreased the lowest levels,there were not significantly differences between B group and C group (p>0.05);②Aftertreatment, the level of serum NSE in the rats treated with xingnao jianshen capsule droppedthan B group at different points times, Although the values of serum NSE of A group werehigher than C group, there were markedly difference between two groups on level of serumNSE at 1d (p<0.05) but there were markedly more difference between A group and B group at1d (p<0.01); A peak of serum NSE emerged at 3d after ICH too, the level of serum NSE of Agroup were high than C group, there were markedly difference between two groups(p<0.05);at 7d, the level of serum NSE of A group start decreased again, there were markedlydifference between A groups and C group (p>0.05);at meantime, the level of serum NSE of Agroup lower than B group, there were markedly difference between three groups(p<0.05);At 14d, the level of serum NSE of A group decreased the most level,there were markedlydifference between three groups (p>0.05);(6),The expression ofNSE mRNA of C groupwere not significant at different points times. The expression of mRNA of B group markedlyincreased at 1d. The peak of the expression of S100B mRNA emerged at 3d. After 3d, itdecreased gradually, At 14d, the level of the expressions of S100B mRNA closed to shamoperation group at corresponding points time; 8),After treatment, the expression of mRNA ofNSE in the rats treated with xingnao jianshen capsule increased markedly than B group at 1d,The peak of the expression of S100B mRNA emerged at 3d, too. it maintained higher level at7d compared with B group and C group. The expression of mRNA of S-100B between threegroup were not significant at 14d. (7),①The positive NSE cells immunoreaction buildedup, the positive nerve cells became less, its protuberance disappeared and the stain dark andlight; at meantime, the quantities of positive cells of B group increased, there were significantly differences between B group and C group (P<0.01); At 3d, the volume of thepositive stain cells increased, but immunoreaction gradually decreased; at meantime, thequantities of positive cells of B group decreased too, there were significantly differencesbetween B group and C group(P<0.01); A peak of immunoreaction emerged at 7d afterICH, the positive nerve cells lysed markedly and the cells velum crash, the cytoplasm wereoverflowed, the space among cells have a lot of positive matters; the stain dark and light, atmeantime, the quantities of positive cells of B group increased significantly too, there weremore significantly differences between B group and C group (P<0.01); The immunoreactionweakend (stain becomed light) from 14d, there were more not significantly differencesbetween B group and C group; Although the quantities of positive cells of B group low littlethan C group at 14d, there were more not significantly differences between B group and Cgroup (p>0.05);②at 1d, the cells form of A group changed no significantly, theimmunoreaction builded up and the quantities of the positive NSE cells increasedsignificantly; There were significantly differences between A group and B group(p<0.05); Apeak of the quantities of positive NSE cells emerged at 3d, there were significantlydifferences between A group and B group(p<0.01); the stain was more dark than B group;the quantities of the positive NSE cells lower than B group at 7d, there were significantlydifferences between A group and B group(p<0.05); the quantities of the positive NSE cellsdecreased the lowest level at 14d, there were not significantly differences between A groupand B group(P>0.05); at meantime, there were not significantly differences between Agroup and B group on the immunoreaction;③The quantities of positive NSE cells of Agroup more higher than C group from 1d to 7d, there were significantly differences between Agroup and C group(p<0.01); the stain darker than C group, but the cell form different withC group, occasionally the cells velum destroyed, the space among cells have some positivematter; The quantities of positive NSE cells of A group decreased the lowest level at 14d,there were not significantly differences between A group and C group (P>0.05).Conclusion: S100 Band NSE are sensitive markers for illness degree and prognosisof acute intracerebral hemorrhage(ICH), the level of serum S100B and NSE and illnessdegree present positive correlation, on prognosis, the level of serum S100B and NSE presentnegative correlation.The mechanism of NSE and S-100B level increase in the blood after ICH is not only due to the leakage from damaged brain cells but also through a highexpression of mRNA and protein. Xingnao Jianshen capsule can adjust the genic expression.①it can restrain the excess hyperplasia and activation of damaged glia cells, and reduce thedisadvantage of damaging nerve cells and exert the advantage of restoring nerve cells.②itcan drop the level of transcription and translation of S100B by reducing strongly the excesshyperplasia and activation of damaged glia cells at forepart, that can reduce the toxic nerveprotein effect of damaged glia cells induced after ICH.③at last phase, it can increaseproperly the level of transcription and translation of S100B, which can exert its protectingeffect on protecting nerve cell.④The experiment indicated: By treatment of XingnaoJianshen capsule, the level of transcription and translation of NSE and S100B stronger thancomparison group, but corresponding result indicated that the effect of xingnao jianshencapsule for S100B stronger than NSE. On the one hand, Xingnao Jianshen capsule can adjustthe glia cell and S100B, which exert the double function of the glia and S100B in the nervecentre system, at meantime, the level of transcription and translation of NSE are adjustedindirectly by above action. On the other hand, Xingnao Jianshen capsule can affect directlythe level of transcription and translation of NSE. The high activation and the energymetabolizing obstacle of nerve cells around the haematoma were improved in times. So thenerve cells around the haematoma correspondingly maintain into an peaceable in-environment,which can decrease the snatch for NSE inner store, so as to reduce the excessive loss of NSE.Meanwhile, Xingnao Jianshen capsule can increase the level of transcription and translationof NSE, At this rate, the produce and consumption of NSE maintain in an relative balanceablestate, Which can improve the status of ischemic and anoxic after ICH in times.
