| ObjectiveDanxiantang have been proved to be effective to the disease of hyperplasia of mammarygland(HMG) by the long-time clinic practice and experiment reseach of proferssor Xu ZhiYin.Danxianpian are composed with Yinyanghuo,Danpi and Kunbu in the modern pharmaceutic technology by drawing up and condensing,with the modern medicaltheory and experimental method,the animals pattern of HMG treated with Danxianpian were detected and observed the shape of breast s,analgestic effect,microcirculation,blood rheology,sexual hormones,pathological changes of breast tissue,and the expression of VEGF,Bcl-2,bax,as well as tissue apoptosis and compared with Rupixiao and Tamoxifen.To research the mechanism of action ofDanxianpian in curing HMG.We hope to provide experiment data for clinical applicationmethodsCombining the Traditional Chinese Medicinetheory to the modern pharmaceutic technology,taking Yinyanghuo 4kg,Danpi 4kg and Kunbu4kg.Paeonol was extracted with steam distillation by several times.The medical water left was heated with Yinyanghuo and Kunbu.the boiling-down water was condensed by three times to thick paste.Commixing the thick paste with Paeonol.Analgestic effect:1.wriggling body method:ICR female mice were randomly divided into 5 groups:the comparative group,the tamoxifen group(0.1mg/kg) the Tramadol group(1mg/kg), the Danxianpian large(5g/kg) dose group and low dose group(5g/kg);the ICR female mice in each group were infused stomach with 0.2ml/10g of various medicine for 3 days.Tthen every mice were injected with 0.3ml acetic acid(1.1%) into abdomen cavity.Record writhing response frequency and the time of writhing response coming.2.Hot plate response:ICR female mice were randomly divided into 3 groups:the comparative group,the Danxianpian large(5g/kg) dose group and low dose group(5g/kg),the ICR female mice in each group were infused stomach with 0.2ml/10g of various medicine for 3 days.To record hot plate sucking feet response frequency in 30',60',90',120'and the time of hot plate response coming.The pattern rats was made by estrone and progesterone injection.Rats were randomly divided into 6 groups:the comparative group,the animalpattern group,the tamoxifen group,the Danxianpian group of high and low dosage.These groups were treated with different drugs.One time one day.After 4 weeks,the rats were taken blood from eyeball without eating food for 12h,then be killed.Observing the shape of breast,blood rheology,sexual hormones etc, E2,P,PRL,pathological changes of breast tissue,and the expression of VEGF,Bcl-2,bax,as well as tissue apoptosis.Result1.The white Paeonol extracted with steam distillation was weighted 42g.The thick paste was 4.012L as equal as traditional and herbal drugs 3g/ml.2.The experiment revealed that Danxianpian can significantly relived the mice writhing response frequency altho without satistic diffierence.And Danxianpian group with high dose extented the time of writhing response coming.Danxianpian reduced mice sucking feet response frequency and lengthened the time of hot plate response coming.3.Danxianpian improved the breast structure of thes HMG and had no difference with tamoxifen(P＞0.05);Effect of both high and low dosage was better than Rupixiao(P＜0.05). Danxianpian meliorated blood hypercoagulability.4.As was shown in the experiment,the high and low dosage of.Danxianpian reduced the content of serum E2 and PRL,and increase the thecontent of serum P(P＜0.01).And Danxianpian including high and low dosage could reduce the expression of tissue ER,PR.There was no difference the Danxianpian group of high dosage from low(P＜0.05) and no distinct difference from the left other groups.5.With observing the tissue of HMG rats treated with Danxianpian,Danxianpian improved remarkly the structure of hyperplasia mammary gland.6.The breasts duct and gland cells of HMG rats contained Bcl-2 high expression(P＜0.01). Danxianpian could reduce the expression of Bcl-2(P＜0.01) and increase the expression of bax(P＜0.05) especially high dose.It demonstrated the Bcl-2 high expression was been restrainted.7.Expression of breast VEGF of HMG rats showed obviously difference from control group (P＜0.01).There was difference the Danxianpian group of high from low dosage(P＜0.05) and no distinct difference from Rupixiao.8.Acute toxicity test:in order to evaluate the satety of Danxianpian in clinical practise,the result showed that Danxianpian might be very safe if it is used as normal dosage.Conclusion:The experiment had proved Danxianpian to be effective to HMG and the entity curative effect better than Rupixiao,be similar to tamoxifen on meliorating blood hypercoagulability and expression of Bcl-2.the reaserch testifid Danxianpian answer to medical treatment on meliorating blood hypercoagulability,increasing blood hemoperfusion;adjusting the disorder of hormone,restrainting expression of hormone rcepector and VEGF,inhibition new bloood vessel;reducing the expression of Bcl-2(P＜0.01) and increase the expression of bax to promote the apoptosis of cells of mammarygland hyperplasia.The experiment revealed cured HMG by relying on various aspect,multi-level and multi link.
