Objective:To confirm the protective effect of moxibustion on acute gastric mucosal injury protection mechanism to explore moxibustion on acute gastric mucosal injury from PAR/TFF mediated ERK signaling pathway.Method:1. SD rats were randomly divided into four groups: control group, model group, moxibustion Zusanli Zhongwan group, moxibustion Zusanli point in the abdominal control group, the experimental rats were first moxibustion Zusanli and a control point in the abdominal pretreatment eight days, and then cause acute gastric mucosal injury model in rats fed with ethanol method, visually gastric mucosal lesion case, the recording gastric mucosal lesion index, light microscopy Morphological changes of gastric mucosa tissue immunohistochemistry assay TGF-a, PCNA, TFF gastric contents in1/2/3, PAR1/4in.2. Compared with the normal group, the model group rat gastric mucosa showed obvious damage, ulcer index was significantly higher (P<0.01), optical microscope, pathological changes were tissue morphology; compared with the model group, moxibustion Zusanli, rat gastric mucosa in the abdominal injury group was significantly less, dramatically reduce ulcer index (P<0.01), a light microscope morphological structure of its normal; and moxibustion Zusanli Zhongwan group phase ratio, moxibustion Zusanli, rat gastric mucosa in the abdominal injury is more serious control point group, lesion index was also significantly increased (P<0.01), a light microscope morphological pathological changes.2. Compared with the normal group, TGF-a, PCNA content in gastric mucosa of rats in model group was significantly higher (P <0.01or P<0.05); compared with the model group, moxibustion Zusanli Zhongwan rats gastric mucosa TGF-a, PCNA content significantly higher (P<0.01or P<0.05); and moxibustion Zusanli Zhongwan group compared to moxibustion Zusanli rat gastric TGF control point in the abdominal group-a, content PCNA significantly lower (P<0.01or P<0.05).3. Compared with the normal group, the model group rat gastric mucosa TFF1, TFF2, TFF3expression were significantly increased (P<0.01or P<0.05); compared with the model group, moxibustion Zusanli in rat gastric abdominal group TFF1, TFF3expression were significantly lower (P<0.01or P<0.05), TFF2although somewhat lower, but compared with the model group was not statistically significant, and moxibustion foot Third, the rat gastric mucosa in the abdominal group TFF1, TFF2, TFF3are at normal levels; moxibustion Zusanli point in rat gastric mucosa in the abdominal control group TFF1, TFF2, TFF3expression and Ai moxibustion Zusanli, showed no significant difference in the abdominal group.4. Compared with the normal group, model group rat gastric mucosa in PAR1, PAR2expression was significantly increased (P <0.01); compared with the model group, moxibustion Zusanli Zhongwan rat gastric mucosa PAR1, PAR2expression was significantly increased (P<0.05); and moxibustion Zusanli Zhongwan group compared to moxibustion Zusanli point in rat gastric mucosa in the abdominal control group PAR2expression was significantly lower (P<0.01), moxibustion Zusanli point in the abdominal control rats gastric mucosa PAR1expression and moxibustion Zusanli, although low compared to the abdominal group, but no statistical significance.5. Compared with the model group, ERK inhibitor group rat gastric mucosa damage index was significantly higher (P<0.01), its morphology was observed under light microscope pathological changes are more severe untreated group; compared with moxibustion acupuncture group moxibustion+ERK inhibitor group rat gastric mucosa damage index was significantly higher (P<0.01), its morphology was observed under light microscope pathological changes are more severe moxibustion acupuncture group.6. Compared with the model group, the rat gastric mucosa ERK inhibitor PCNA expression was significantly lower (P<0.05); compared with moxibustion acupuncture group, moxibustion+rat gastric mucosa PCNA ERK inhibitor group The expression was also significantly lower (P<0.05).7. Compared with the model group, the expression in rat gastric mucosa moxibustion acupuncture group, the submandibular gland, the hypothalamus ERK1/2m RNA was significantly increased (P<0.01 or P<0.05), ERK inhibitor of gastric mucosa, submandibular gland, the hypothalamus in the expression of ERK1/2m RNA was significantly lower (P<0.01or P<0.05); compared with moxibustion acupuncture group, moxibustion+rat gastric mucosa ERK inhibitor, submandibular gland, the hypothalamus ERK1/2m RNA expression were significantly lower (P<0.01or P<0.05).Conclusion:1. Moxibustion pretreatment on ethanol-induced acute gastric mucosal injury stomach has a protective effect, and has certain specific points.2. Moxibustion can increase acute gastric mucosal injury in rats TGF-a expression in gastric mucosa, PGNA, and promote proliferation and restoring of gastric mucosa.3. Moxibustion pretreatment on acute gastric mucosal injury in rats with gastric mucosa PAR, the expression of TFF related.4. PAR/TFF mediated ERK signaling pathway is one of the mechanisms of moxibustion protecting acute gastric mucosal injury.
