Objective:To observe the effects of acupuncturing at Neiguan points (PC-6) on morphology, function and the expression of inflammatory cytokines in isopropyladrenaline induced myocardial hypertrophy model mice.Method:Experimental animals and grouping:SPF male C57BL6 mice were randomly divided into model group, acupuncture group, acupuncture-control group, p-control group and control group,20 mice for each group. Disposition of each group:Model group:subcutaneous injection of isopropyl adrenaline (15 mg/kg) on the back of the mice once-daily for eight days to establish the myocardial hypertrophy mice model. Acupuncture group:acupuncturing at the Neiguan points of anesthetized mice for 15 min,30 min before injecting isopropyl adrenaline; Acupuncture-control group:acupuncturing at the tail roots of the mice instead of the Neiguan points with the same schedule as the acupuncture group; P-control group:atenolol (10 mg/kg) was given by gavage once-daily for 8 days,30min before injecting isopropyladrenaline; Control group:subcutaneous injection of normal saline (1 mg/kg) on the back of the mice once-daily for eight days. ECG and cardiac function collection:at the 9th day, standard 12-lead electrocardiogram of the mice were recorded after anesthesia and the left ventricular anterior wall thickness (LVAWD), left ventricular end-diastolic front wall thickness (LVAWS), end systolic left ventricular posterior wall thickness (LVPWD), left ventricular end-diastolic back wall thickness (LVPWS), end systolic ejection fraction (EF)% and calculate the left ventricular short axis shortening rate% (FS) were calculated by the small animals high frequency color ultrasound instrument. Heart weight/tibia length ratio:heart quality (HW) was weighted and the tibia length were measured at the 9th day of the experiment. Mortality of the mice: Death of the mice was noted during the whole experiment for the further analysis. Myocardial morphology:HE staining was used respectively to ob-serve the change of myocardial cell, and Masson staining was used to observe the degree of myocardial fibrosis, observed by transmission electron mi- croscopy groups of the changes of the ultrastructure of myocardial cells in mice. Expression of myocardial ANP and TNF alpha were detected by im-munofluorescence and western blot.Result:1. The general activity of the mice in model group and the acu-puncture-control group was obviously lower than that in the control group, with obviously breathing mice, acupuncture group and drug group mice ob-viously better than that in the model group.2. Mice mortality analysis:the mortality of mice in model group and the ac-upuncture point group was obviously higher than that of control group, and acupuncture and p-control groups can be decreased by reducing the mortality rate in mice.3. Echocardiography analysis:compared with the control group, model group and the acupuncture-control group in isopropyl adrenaline subcutaneous in-jection eight days later showed myocardial hypertrophy and heart function. With LVPWD, LVPWS, LVAWS increases with significant statistical signifi-cance (p< 0.05), and EF and FS value does not change significantly between groups, no statistically significant difference (p> 0.05).And compared with model group, acupuncture group and p-control group showed antagonism isopropyl myocardial hypertrophy caused by adrenaline process, its main performance for LVPWD, LVPWS significantly lower than the model group, significant difference, statistically significant (p< 0.05).4. Heart weight/Tibia length ratio analysis:compared with model group, acupuncture, p-control group and the control group mouse heart weight/tibial length ratio was significantly lower, (p< 0.01), significant difference statis-tically significant; Compared with the control group, model group and the acupuncture-control group mouse heart weight/tibia length ratio increased significantly,(p<0.01), significant difference statistically significant. And p-control and acupuncture group heart weight/tibia length ratio (p> 0.05), no statistically significant difference.5. Morphological changes of myocardial tissue:HE staining and the projec- tion electron microscope shows that acupuncture and drug group can im-prove myocardial cells and the pathological changes of muscle fibers caused by isopropyl adrenaline, Masson staining:compared with the control group, model group and the p-acupuncture group shows obviously myocardial fibers fibrosis, p< 0.05, significant difference statistically significant. Compared with model group, acupuncture group and p-control group significantly re-duced myocardial fibrosis degree, P< 0.01, significant difference statistically significant.6. The expression of mouse myocardium ANP analysis:by immunofluores-cence and western blot analysis as compared with model group, acupuncture group and p-control group can significantly reduce the expression of ANP in mouse myocardium, significant difference statistically significant, but the acupuncture-control group does not reduce the expression of ANP in mouse myocardium, there was no significant difference with model group.7. compared with the control group, model group and the acupuncture-control group had a significantly speed up heart rate of mice, p< 0.01, significant difference statistically significant; And compared with model group, acu-puncture group and p-control group can significantly reduce the heart rate of mice, p< 0.01, significant difference statistically significant; When compared to heart rate of p-control and acupuncture group, p> 0.05, no statistically significant difference.8 analysis of TNF alpha in myocardial tissue:the study found that compared with model group, acupuncture group and p-control group can significantly reduce the expression of TNF alpha in mouse myocardial, significant differ-ence statistically significant, but the acupuncture-control group does not re-duce the expression of TNF alpha, there was no significant difference with model group.Conclusion:1. Acupuncture at Neiguan points can adversely affect isopropyladrenaline to myocardial hypertrophy in mice morphological and functional changes.2. Acupuncture at Neiguan points can reduce myocardial hypertrophy com-pensatory periodic heart rate in mice.3. Acupuncture at Neiguan points can decrease myocardial hypertrophy my-ocardium ANP and the expression of TNF alpha in mice.
