The Research Of Shuxiong Soft Capsules

Shuxiong Soft Capsule is made of Radix notoginseng, Flos carthmai and Rhizoma chuanxiong, which has the effects of promoting blood circulation, removing blood stasis and relieving pain, used in blood stasis obstruction, oppressive pain in the chest and heart, surgical trauma, swelling and pain caused by blood stasis; coronary heart disease, angina pectons, cardiac arrhythmia, soft tissue injury and so on. The product was made into soft capsule based on clinical application, by choosing and proving of prescription and dosage form. This subject mainly deal with: (1) establishment of a method for determining the content of principal component; (2) extraction technology of crude drug; (3) preparation technology of soft capsule; (4) quality standard and stability of preparation; (5) pharmacodynamics research.In this study, the method of TLC was used to identify ginsenoside Rg₁ and ferulic acid in herbs and shuxiong soft capsules. Compared with controls and herbal controls, the sample had the pot of same color in the corresponding position. Negative sample hadn't any interference. The method of HPLC was built up to determine the content of two main ingredients. The two methods were accurate, reliable and repeatable.According to the property of herbs, Radix notoginseng, Rhizoma chuanxiong and Flos carthmai were extracted and refined in different ways. (1) Researching on extraction technology of Radix notoginseng. It was extracted by percolating with alcohol. Experiment was designed according to L₉(3⁴) orthogonal experiment which containing four factors—alcoholic consistency, alcoholic amount, extraction time and frequency. Three levels were chosen in every factor, and the concentration of Ginsenoside Rg₁ in Radix notoginseng and dry paste ratio were regarded as evaluation index to optimizing extraction technology. The results showed that the best extraction technology was that Radix notoginseng rough powder was soaked for 12 hours by 8 folds 60% alcohol, then was percaolated with flow rate at 5ml/min. (2) Boiling with water was used to extract Rhizoma chuanxiong. We did experiment according to L₉(3³) orthogonal experiment containing four factors—the amount of water, extraction time and frequency .
