Xingnaojing injection is one of the patent medicine affirmed by Ministry of Public Health for the clinic first aid. It is also one of the predominant products of Shanhe Pharmaceutical Co., Ltd. with the good economic and social performances. But the traditional processing technique, double steam distillation (SD), exists such disadvantage as the low utilization ratio of raw medicine materials, high production cost and so on. With the continuously rising in Muschus market price, the study on improving the medicinal materials utilization ratio especially for Muschus is very important for the development of the corporation by reformation the lag craft. Considering the particularity of the natural medicine, the paper studied the feasibility using supercritical fluid extraction (SFE) method to extract the compound Xingnaojing.The SFE for the volatile oil (SFE oil) in Curcuma wenyujin was studied by orthogonal test. The analysis of the volatile compounds in the SFE oil and the steam-distilled oil was carried out by the gas chromatography-mass spectrometry method. The result indicated that extraction time was the first influence factor for the oil extraction ratio. The optimum SFE conditions for extraction were found as follows: extraction temperature 55℃, extraction pressure 35 MPa, extraction time 240 minutes. Eleven compounds were first found in Curcuma wenyujin, such as nonanal, aromadendrene oxid, ledene, ledene oxide, calarenepoxide, junipene, isoaromadendrene epoxide and so on. Comparing with SD, SFE showed advantages, including high extraction yield, short processing period, high contents of active components witch the rate of the elemene, curdione, Curcumol and the germacrone increased 32%,23%,105% and 15% respectively.The SFE for the volatile oils in Gardenia jasminoides Ellis were also studied by orthogonal test, and the analysis of the volatile compounds obtained from the SFE oil and the steam-distilled oil by the gas chromatography-mass spectrometry method was done. The pressure had significant influence on extraction oil yield, while the influence of temperature was in the mediacy and the influence of extraction time was smaller. The optimum conditions were: extraction temperature 55℃, extraction pressure 35 MPa, extraction time 120 minutes. The SFE oils consist of linoleic acid, palmitic acid and trans, trans-2,4-decadienal, but the main content of the SD oils was trans, trans-2,4-decadienal.Application the uniform design method, with the relative rate of muscone and volatile compounds as the guideline for the experiment, established the mathematics regression model and optimize the SFE condition for the volatile compound from Moschus, the analysis results indicate at the condition of extraction temperature 35.0℃, extraction pressure 25.0 MPa, extraction time 150.0 minutes the muscone rate was 21.2674 mg / g raw medicinal materials, the volatile compounds rate was 16.29%.
