Study of active ingredients in traditional Chinese medicine is one of the most and important problems during its modernization, and basic research on effects and mechanism of active ingredients has important theoretical and practical meaning. Flavonoids and ginsenosides have many kinds of pharmacological and physiological properties, and will be an exciting research field. Stauntonia chinensis DC, containing ginsenosides, proteins, flavonoids and suchlike, is good for health protection and cosmetology. Stauntonia chinensis DC extract has been used for relieving the pain, calmness and has few side effects; Stauntonia chinensis DC extract prepared by me has been proved to be anticancer. But basic research about it develops not fast enough, and blocks its practical application. In this paper, several extraction methods of active ingredients from Stauntonia chinensis DC were compared, and analytical methods were also studied with UV, HPLC and electrochemical techniques.Firstly, soxhlet extraction was compared with sonication, and we choose the latter, because it has more advantageous, for example, saving time, higher extraction efficiency and the samples need not to be heated. Under optimized experiment conditions, flavonoids and ginsenosides were extracted from Stauntonia chinensis DC by sonication, and the content of them were determined by UV. These techniques will make good foundation for the further use of Stauntonia chinensis DC in clinical medicine.Secondly, RP-HPLC was used for analysis of oleanolic acid in Stauntonia chinensis DC, and reproducibility of the method was studied. Then two-phase extraction was adopted to get the active compounds from the ethanol extract, and the fraction is analyzed by HPLC-UV.The information of UV spectra of the extracts was obtained and the qualitative analysis was discussed primarily with HPLC-UV results. The electrochemical behaviors of the ethyl acetate extract and the n-butyl alcohol extract at a glassy carbon (GC) electrode were also investigated by cyclic voltammetry. The results showed that the electrode process of the two types of extracts at the GC electrode is a quasi-reversible process with absorption and diffusion,and the results are good for finding a fast, simple and convenient electrochemical analysis technique. The antioxidant activity of the two types of extracts was discussed with the results of CV and DPV.Finally, MTT assay was adopted to determine the effects of Stauntonia chinensis DC extracts on cancer cell line K562 and the main active ingredients were discussed primarily.
