Nano-technology, which is a new technique born in 80's, 20 centuries, will be one of important contents of technological industry revolution in 21 centuries. Scanning Tunneling Microscope(STM), a new type of microscope with high resolution capability based on quantum tunnel effect, following the first generation-optical microscope and the second-electronic microscope, is the third generation microscope. As a kind of new surface analytical instrument, STM is mainly applied in the research of the surface structures and properties of conductive materials in many fields. With its atom-scaled resolution capability, STM may observe the geometry distribution and distributive density of the atoms or molecules on the surface of the sample, which brings about nano-medicine, nano-biology, nano-material science and nano-mechanics. Meanwhile STM is gaining ever-increasing popularity in its application with the coming of nano-technology. As a result many developed countries regard it as the main direction of technology development. Besides the stabilization of the instrument itself, the quality of its pictures, the simplicity of its operation, the expansion of its application field and its industrialization have also drawn more and more concerns.As a new instrument, STM is still a subject in need of further research. The research in this paper was supported by the National "985" Key Subject-construction Program. The major work in this paper is as following:In this paper, the current situation of STM inside and outside the country, the advantages and disadvantages of STM, and the main application field of STM were introduced. The principle of STM's operation, the electronic tunnel effect phenomenon and the tunnel current were also introduced.The key techniques of STM IPC-205B were shown as following: the design of the micro-displacement transmission system with wide scope and high speed; the physical mechanism and vibration-proof measures which greatly influence the theoretical precision of STM; the basic design theory of scanning system and data collection system; the methods of the intelligent control of scanning voltage and the logarithm contraction of tunnel current.The process of making probe and the three-dimension reconstruction and disposal of the scanning picture were researched.By STM IPC-205B , the scanning tunneling spectroscopy of calcium carbonate surface the surface shape of graphite, nickel nanocrystal, zinc oxide crystal were observed, and the high quality pictures of sample surface shape were attained.
