By polyacrylamide gel electrophoresis (PAGE) with discontinuous vertical plate, isoenzymes of Esterase (Est) and Lactate Dehydrogenase (LDH) were studied in FI generation (LW & XWS-?, which was composed of the hybrid offspring of Wannan-Spotted Swine (WS) and Large-White Swine (LW). By means of ultraviolet spectrophotometer, the enzyme activities of LDH, amylase (Amy), alkaline phosphase (Akp), cholinesterase (ChE), glutamatepyruvate transaminase (GPT), glutamicoxalacetic transaminase (GOT) and creatine phosphate kinase (CPK) in plasma were estimated. In addition, the relationship between those isoenzymes and productive traits were analyzed by Least Squares Model (LSM) and Multivariate Regression Analysis.The results showed that the plasma Est Locus controlled by two alleles had three genotypes. Through chi-square test (x2-test), three polymorphic genotypes of the gene locus were in Hardy-Weinberg equilibrium. Gene frequencies at Est locus were EstA(0.5527) & EstB (0.4473) in LWXWS, and the EstA was the dominant gene. Genotype frequencies were ?1(0.3158), ?1(0.4737) & EstBB(0.2105), and EsfAB was the dominant one.Genotype Est had positive effect on nearly all the traits, except heart weight. Analyzing the relationship between the scanning value of plasma Est and traits, we found that both Est4 and Ests had positive effect on morphology, carcass and tissue organ traits. At the same time, we found that Est] and Est2 had the similar effect on meat quality traits.Plasma LDH in LWX WS swine also controlled by two alleles of two loci had four genotypes, namely LDHi, LDH2, LDH3 and LDH? but no LDH5. LDH2had the positive effect on almost all traits. On the contrary, LDHa had the negative effect.The Est electrophoretic spectrum in tissue and organ were complicated. According to phenotypes, we defined the bands of Est isoenzyrne from anode to cathode as Esti, Esta, æ™»? Estio- Distribution of Est isoenzyme showed speciality, because different tissues or organs had different enzyme bands, the number ranging from 3 to 10 and the relative enzyme activities in different tissues or organs were also different. The activities of Esti and Est2 in plasma and spleen were higher than in other tissues. Est3, its activity to be highest of nine tissues or organs, was one of the three bands of lung. The activity of Estjwas less than 11.40 percent in all nine tissues or organs. The activity of Estsof heart (53.05%) differed significantly with other tissues at P<0.01. The relative activity of Est6 (35.77%), Est7 (53.53%) and Estg (17.13%) were the highest in stomach, large intestin and in muscle, respectively. Estg was expressed in spleen, muscle and little intestin, but its activity was less than 13.85 percent. Moreover, Estio was expressed in muscle and little intestin. Significant phenotypic correlation existed between Est in tissues or organs and morphology traits, and with carcass traits, but showing no distinct rule.Subunits of LDH in different tissues, as above mentioned, had different characteristics, including classes, relative contents, activities and ratios, and we deduced that it was because of the speciality of gene expression. The activities of LDHi of plasma, heart, stomach, kidney, ovary and womb were higher than that of other tissues. Subunit ratios of H to M in those tissues were more than 3:1, and H was dominant subunit. However, the activities of LDHs mainly showed in muscle, Subunits ratios of H to M of which were 0.25:1, and this time, M subunit was the dominant one. Expression activity of tissue LDH had larger effect on meat quality and tissue organ traits than on morphology and carcass traits.Significant phenotypic positive correlations existed in morphology traits, carcasstraits and between them. Moreover, these two kinds of traits had the trend of significant phenotypic correlations with meat quality traits and tissue organ traits.Plasma enzyme activities changed with the day-age in Wannan-spotted swine.
