The anthers of loquat"DaWuXing" were used for explants in the study. The main objectives of the study were to investigate the factors affecting callus induction and to find out the optimium culture media especially hormone supplements. The results were as follows:(1) The suitable period for anther culture was middle October(2) Anthers from young bearing trees were more advantageous for the induction of callus than from older bearing trees.(3) The cold pretreatment(5℃,3~7days) couldn't effectively raise the callus induction frequency in anther culture. The frequency of callus induction was highest when the anthers were pretreated at 5℃for 3days.(4) Sucrose was advantageous for the induction of callus and the optimum concentration of sucrose for callus induction was 30g/L. Starch was not advantageous for the induction of callus.(5) The optimum medium for callus induction was MS supplemented with 6-BA2.0mg/L and 2,4-D1.0mg/L, and frequency of callus induction was as high as 90.00%. 2,4-D was most important of all the hormonal regulants including 6-BA and 2,4-D.The frequency of callus induction, 87.62%, on medium with both 6-BA and KT was significantly higher than that on medium with 6-BA, KT and ZT respectively.(6) The best multiplication media were MS supplemented with 6-BA1.0mg/L, KT1.0mg/L and IBA0.8mg/L,and MS supplemented with NAA1.0mg/L and BA0.25mg/L.(7)Culturing anther in dark could raise the frequency of callus induction.
