Construction of in vitro culture system, rejuvenation and renewal of aseptic plantlets, and plantlet transplant of anthurium (Anthurium andraeanum) cultivars 'Jolanba', AI and BI were studied. In vitro culture system of 'Jolanba' was established. Endogenous phytohormones levels and other parameters of three calli of 'Jolanba' with observably different redifferentiative behaviors were investigated to discuss physiological and biochemical mechanism of redifferentiation in anthurium. In vitro morphogenesis pathway of anthurium and anatomic differences among eight calli from different culture stages or culture media were discussed. The correlation between genetic relationship and in vitro regeneration capability of anthurium cultivars was approached by using RAPD technique. The results went as followed.The difference of morphogenesis in vitro among Anthurium cultivars was significant. The base part of fully unfolded deep red leaf and just turning green leaf was better for callus induction. The medium components combination of l/2macro+1.0 mg/L BA+0.1 mg/L 2,4-D+3% sucrose and inoculation with leaf face upwards were appropriate. As for shoot induction, MS+0.5 mg/L BA+0.1 mg/L NAA was the best combination in this experiment.Concentration of exogenous phytohormone should be decreased until to 0 progressively during in vitro culture and activated charcoal had a remarkable effect on rejuvenation of regenerated plantlet. Shoot tips of plantlet in vitro were best for callus induction of renewal culture and l/2macro +1.0 mg/L BA+0.1 mg/L 2,4-D was preferable. Shoots were induced subsequently in MS+0.01mg/L TDZ.Regenerated plantlet with height of about 3cm, 2-3 roots of 0.5-1.0cm long and maximal leaf of over 0.8cm wide was perfect for transplant. Perlite or combination of peat and perlite with a ratio of 2:1 was used for transplant.High content of soluble protein favored shoot differentiation, and high content of soluble saccharide favored shoot growth. There was an obvious positive correlation between activities of POD & SOD and number of differentiated shoots. So was between GA(1+3)/ABA, as well as ZRs/ABA, and shoot height. There was also relation between IAA/ABA and rhizogenesis.The main pathway of in vitro morphogenesis of Anthurium 'Jolanba' was indirect organgenesis, and embryogenesis was also found. Eight calli differed in type of nodule and number & distributing of meristem.There was correlation between regeneration capability and genetic relationship.The following researchs in this thesis has not been reported yet: (1)renewal of asepticplantlet and effect of TDZ on shoot differentiation during this course;(2)approach onphysiological & biochemical mechanisms and anatomic characteristics of Anthuriumredifferentiation; (3)discussion of correlation between genetic relationship and regenerationcapability.
