The explants from the embryo of mature seeds and bud-stem as well as leaf segments of Sorbus folgneriwere used to establish the technique system of tissue culture and rapid propagation. By means of the orthogonal experiment design, analysis of variance and multiple comparisons, the best medium and culture procedure of each phase were analyzed. The experiments showed that:(1)Using the embryo of mature seeds as explants, the optimal initiation medium for shoot regeneration was 1/2MS+2. Omg/L 6-BA+1.5mg/L NAA, the inducement rate could be up to 90%.(2) Using the bud-stem segments as explants and picked it in spring, the best initiation medium was 1/2MS+2.0mg/L 6-BA+l. 5mg/L NAA and the inducement rate could be up to 90%.(3)The best initiation medium for bud multiplication was 1/2MS+1.5mg/L 6-BA+l. Omg/L NAA, and the multiplication rate could be up to 4.(4) Using the young leaves of Sorbus folgneri as explants to induce callus, the best medium was 1/2MS+ 0. 5mg/L 6-BA+0.1 mg/L NAA+ 0. 5mg/L 2, 4-D, and the inducement rate could be up to 90%.(5)The best medium for callus induce cluster buds was 1. Omg/L KT+1.0mg/L 6-BA+ 1.5 mg/L NAA+1/2MS , and the inducing rate could be up to 90%.(6)The best medium for the rooting of shoots was 1/2MS +1. 0 mg/L IBA+0. 1mg/L NAA +0.5 mg/L AC+10g/L Sugar, in which the rooting rate could reach up to 75%.(7)The survival rate could be up to 80% when the rooted test tube plants were transplanted into the medium with 1/3 vermiculite and 1/3 ruby mica and 1/3 turf.
