Tissue culture technology is an important way of rapid propagation for Kiwifruit.At present,the reports of Kiwifruit tissue culture are numerous.However,there are few studies on the new varieties of Kiwifruit,such as RB-1 ? MHT-17 and MHT-19.This dissertation carries on an experiment study of three varieties of Kiwifruits from two aspects--one exploring the medium formula suitable for the three Kiwifruit varieties in the tissue culture stage;the other exploring the domestication and transplanting technology of Kiwifruit tissue-culture container seedlings in the greenhouse.This dissertation is designed to shorten the seedling period of kiwi tissue-culture seedling and provide a theoretical and practical basis for an efficient,stable system of rapid propagation technology.The main findings are as follows:1.The sterilization time and stem tip size have impacts on the asepsis explants: in the 16 treatment groups,the samples are processed in the 70% medical alcohol for 40 s,and flushed in sterilized water three times.After that.soaked in 0.1% HgCl2 8 minutes,stem tip size 0.3 mm,the survival rate and starting rate reach to the highest,the survival rate of90.9%,starting rate of 85.5% while the pollution rate of 25.4%.2.Under the condition of darkness,S6(MS+0.1mg·L-1NAA+1.0 mg·L-1ZT)and S7(MS+0.1mg·L-1NAA+1.5 mg·L-1ZT)combination has no significant difference,which medium induced stem section forms callus of the best effect with the induction rate of94.4%?93.4%,the callus pale green,transparent as well as a better growth status;3.In inducing callus from bud formation,ZT has a distinct high efficiency.S7(MS +0.1 mg·L-1NAA + 1.5 mg·L-1ZT)has the highest induction rate of 91.8% as well as a propagation coefficient of 7.3 times higher;4.In the induction of lateral bud germination,M3(MS + 0.1 mg·L-1NAA + 1.0mg·L-16-BA),with the corresponding axillary bud germination rate 88.2%,45.5% higher than that of the control group on average,the highest and axillary bud growth robust and dark green.5.In the rooting culture of Kiwifruit container seedlings,compared with the control group,different concentrations of IBA treatment exert obvious influences on the adventitious bud rooting where(1/2MS + 1.0 mg·L-1IBA)culture medium makes fruit grow the best with rooting rate of 76.3%,the average rooting number of 6.0 roots,root length of 2.62 cm,color white,strain height up to 2.6 cm,rooting rate 57.3% higher than that of the control group.6.Adding appropriate concentration of the activated carbon to the rooting medium(1/2 ms +1.0 mg · L-1IBA),is instrumental in reducing the pollution rate of container seedlings.The most appropriate being 3 g·L-1 content.Survival rate is 92.32%,increased by 19.5%.Rooting rate is 95.4%,decreased by 16.8%.7.In October 2014,transplanting experiment demonstrates that the survival rate is highest,about 92.0%,on the seventh day,when seeding is high 4cm.8.Different concentrations of carbendazim and processing time have a significant impact on the survival rate of transplanted tissue-culture seeding.There were huge differences between he treatment group and control group ck,where the x5?carbendazim1500 times liquid,with 60 s treatment?processing leads to the highest transplanting survival rate of 93.0%,19.6% higher than ck.9.Under the same culture management,11 kinds of matrixes have effects on survival rate of the tree transplanted Kiwifruit varieties,where y7?pure coconut chaff?,c4?peat,vermiculite volume = 2:1?matrixes lead to high transplanting survival rate of the three transplanted Kiwifruit,an average of about 90%.10.Transplanted 2 months later,y7,c4 matrix,the three varieties of Kiwifruit are surveyed and measured their root growth situation.The results show that the y7 matrix is instrumental in root elongation and root activity enhancement and advantageous to the longitudinal growth of the new shoot;c4 matrix is beneficial to the root induction and enlargement and favorable to the lateral growth of stem and leaf chlorophyll synthesis.
