Male sterility is very popular in high plant, it is often used as female parent to produce hybridism F1 without getting rid of androecium, which can decrease the production cost and enhance the homozygosis of crossbreed. So male sterility have been researched popularly by Domestic and abroad scholar for many years. In this paper, we research the male sterile line "zhengxing1hao" and fertile line in eggplant from the following aspects.一, Tissue culture research on male sterile line in eggplantThe result indicate that culli induction have something to do with the concentration of 2,4-D and 6-BA, The best concentration of 2.4-D and 6-BA is 0.5mg/L and 1mg/L, respectively.if the concentration of 2,4-D and 6-BA is too high, the calli can not proliferate, the colour of the calli turn brown and then the calli will die. By culture , A lot of adventitious buds differentiate if the concentration of 6-BA and IAA is 3-5mg/L and 0.5-lmg/L,respenctivly. the best concentration of 6-BA and IAA is 5mg/L and 0.5mg/L,respectively. The adventitious buds grow slowly in the culture medium for buds growing medium. The ability of diggerentiating decrease when eggplant become sterile.二, molecular analysis of male sterile line and fertile line ineggplantThe total DNA from male sterile line(MS) and its fertile line (CK) in eggplant were screened primarily by 59 random sequence 10 mer primers,and from which 5 primers, amplifying reproducibly polymorphic bands between CMS and CK each time, were indentified. Then the 5 primers were further used to amplify their total DNA ,nucleus DNA and mitochiondrial DNA(mtDNA), respectively. And 12,6 and 9 polymorphic fragments ,which are different between A and B, were produced when total DNA, nucleus DNA and mtDNA were used as template, respectively. Among these 6 fragments which were produced when the nucleus DAN were used as template , there are 4 occurred in male sterile line and 2 in fertile line. These resultsindicate it is different in nucleus DNA between MS line and fertility line. Among these 9 fragments which were produced when the nucleus DAN were used as template, there are 5 occurred in male sterile line and 3 in fertility line. These results indicate it is different in mtDNA between MS and CK. The result indicate that the sterility is induced by nucleus DNA and mtDNA.三, Biochemistry analysis of male sterile line and fertile linein eggplantwe extract the protein from male sterile line and fertile line in eggplant and analyze them by SDS-PAGE and Two-dimensional electrophoresis. The result indicate that there is obvious difference between the protein of male sterile line and fertile line. These are 50KD , 40KD and 20KD lanes in the protein of male sterile line, but not in the protein of fertile line. There are two kinds of difference between the protein of male sterile line and fertile line by Two-dimensional electrophoresis. One is that there are some protein point in male sterile line but not in fertile line. The other is that there are some protein point in fertile line but not in sterile line.
