Experimental Study On Chinese Traditional Medicine And Ribosome Inactivating Protein Resisting Virus

This research consist of three parts and mainly include Chinese traditional medicine resists NDV and AIV and ribosome inactivating protein(RIP) resists NDV, AIV and FPV.The first part is about experimental study on Chinese traditional medicine resisting NDV. By embryo and the cell culture in vitro, the cytotoxic influence and antivirus mechanism of action of the eleven Chinese herbal medicines which were Andrographis paniculata Nees, Isatis root, Isatis leaf, Flos lonicerae, Radix Scutellariae, Dry opteris crassirhizoma Nakai, Rheum officinale Baill, Polygonum cuspidatum Sieb and so on, were systematically studied in three ways which were adding after medicine and virus acted for a while, adding virus before adding medicine and adding medicine before adding virus. Through directly inactivated action, treatment and preventive action of Chinese traditional medicine to three different NDV strains, Flos lonicerae, Radix Scutellariae, Rheum officinale Baill and Polygonum cuspidatum Sieb are proved to be able to prevent and cure NDV better, which were then main parts of complex prescription. Then by chicken embryo, CEF and animals, complex prescription were tested whether it can resist NDV better.The result shows that it can resist NDV better at chicken embryo and CEF. By chicken embryo, minimum concentration of medicine of directly inactivated test, treatment test and preventive test is 7.5mg/ml, 62.5mg/mland 31.25mg/ml;By CEF, minimum concentration of resiting NDVF₄₈E₈ and moderate virulence strain is 10mg/ml, 50mg/ml and 25mg/ml. However,the result of complex prescriptionpreventing and curing NDVF48Ej$ is not ideal.The second part is about experimental study on Chinese traditional medicine resisting AIV. The experimental methods are similar to ones above. By chicken embryo and CEF, the cytotoxic influence , directly inactivated action treatment and preventive action of the ten Chinese herbal medicines which were Andrographis paniculata Nees^ Isatis robU Isatis lea^ Flos Lonicerae> Radix Scutellariae^ Fructus Forsythiae^ Dryopteris crassirhizoma Nakai> Rhizoma Coptidis and so on.Then Dryopteris crassirhizoma Nakai, Radix Astragali-, Rhizoma Astragalin Foliom Isatiois and Fructus Forsythiae are proved to be valid and are the main parts of complex prescription of resisting AIV. This part is only about the study in vitro, but the result of the study in vitro is not identical to ones of the study of animals. The animal experimental study on complex prescription resisting AIV needs to be studied farther.The third part is about the experimental study of RIP risisting virus in vitro. RIP is a kind of toxin plant protein in which extensively lives in the body of higher plants and controls ribosome' s function. Beside it can control protein' s combination, it has lots of biological reactivity as resisting giving birth and tumor and controlling HIV. At first, RIP is isolated from seeds of bitter melon. The result of SDS-PAGE indicates that there are lots of R*IP in the abstraction liquid. Then Then we study the antiviral action of RIP through the cell of CEF and chicken embryos. The results show that RIP can resist NDVF4gEg, AIV and FPV.
