This experiment researched the effects of substrate's volume and density on Pleurotus ostreatus mycelium's germination, its mycelium's growth rate, extracellular enzymes activity, biological efficiency and Pleurotus ostreatus' quality. The experimental date was dealt with and analyzed statistically with variance analysis and the shortest significant ranges, expounded the variance significance that substrate's volume and density brought. In the end the best treatment and the suitable density were put forward to provide some reference basis for Pleurotus ostreatus' production.The results showed that Mycelium's germination was effected by volume and density, and less density mycelium germinates better, stronger, more vigorous and faster. The difference between volumes is less significant while the difference between density is the most significant. The difference between interaction effect from volume and density is also the most significant. The activity of cellulase was influenced by volume and density, the difference is not significant between volume, but the difference between density and between interaction effect from volume and density are all most significant, treatment A1B1's cellulase activity is the strongest; The activity of laccase, the difference is the most significant between volumes, the difference is also the most significant between density, but the difference is not significant between interaction effect from volume and density, treatment A1B1 and A2B1's laccase activities are both the strongest (equal); The activity of polyphend oxidase, the difference are all the most significant between volume, between density and between interaction effect from volume and density, treatment A1B1's polyphened oxidase activity is the strongest. Pleurotus ostreatus' biological efficiency is also affected by volume and density: Under equal density, the larger volume is, the higher biological efficiency is. Treatment A3B1's efficiency is the highest 112.7%. There are no significant differences in the effects that different volume and density have on Pleurotus ostreatus' quality. In view of comprehensive effects, B1(0.42g/cm3) is the suitable density, but A3B1 (4900cm3 , 0.42g/cm3)is the best treatment in consideration of biological efficiency.
