The shoot tip and stem segment of 'Gui Fei Zao' were used as explants in study of organ culture to establish a rapid technique system of micropropagation. By means of the orthogonal experiment design, analysis of variance and multiple comparisons, the best medium and culture procedure of each phase were analyzed. The results were as follows:l)The explants from inoculated seedlings were the best among inoculated seedlings, infancy disparted seedlings and mature trees ,which had both low contaminated rate, better life and could gain the mature trees fastly.2)The middle part in the same plant was the best part of gaining explants, which had low contaminated rate and better sprouting effect.3)June was the best period of picking the explants in the test and had the best sprouting effect.4)The best initiation medium was H+BA1.5mg/L+IBA0.3mg/L,and the inducement rate could be up to 71.93%.5)The best medium of multiplication of clustered buds was 2MS+BA2.0mg/L+ KT0.3mg/L+IBA0.4mg/L,and the average rate reached 9.0 upwards per generation.6)The suitable medium for the single plantlet growth was l/2MS+KT0.5mg/L+ NAA0.05mg/L.After fostering, the Exterior characters of the single plantlet, such as hight, leaf colour, leaf form and so on, were very well.7)The best medium for the rooting of plantlets was l/8MS+IBA1.0mg/L+NAA0. 3mg/L+sucrosel0g/L,in which the rooting rate could up to 81.83%.8)The best transplantion period was about sixty days.Then the plantlets growed very well, which had much thick and long root with fibres. Meanwhile the leaveswere bright green and unfolded , which could have sufficient photosynthesis to improve the survival rate of plantlets.9)For nursling of plantlets, the soil of the rotten leaves was the best materials, and the survival rate could be up to 80%.
