A marker-phenotype correlative analysis of 7 phenotype wool traits of 129individuals of 6 sires in a Liangshan Semi-wool Sheep nucleus breeding population wasmade by adopting thirteen microsatellite markers on chromosome 3 and two microsatellitemarkers on chromosome X in sheep genome, binding polymerase chain reaction (PCR)and multiplex gel electrophoresis followed by silver staining. The 7 phenotype traitswere:staple length,staple length at 18 months,staple length at 30 months,wool yield at 18months,wool yield at 30 months,wool fibre diameter at 18 months,wool fibre diameter at30 months.The conclusion of phenotype correlative analysis of 7 wool traits is that:staple lengthat 18 months is extreme significantly correlated with staple length at 30 months(p<0.01);wool yield at 18 months is extreme significantly correlated with wool yield at 30 months(p<0.01);wool fibre diameter at 18 months is significantly correlated negatively withstaple length (0.01<P<0.05), is significantly correlated with wool fibre diameter at 30months (0.01<P<0.05); wool fibre diameter at 30 months is extreme significantlycorrelated negatively with wool yield at 18 months (P<0.01).The results of this experiment are as following:the average number of alleles for 15microsatellite markers is 8 (from 5 to 11); the average polymorphic information is 0.754(from 0.252 to 0.859);the average heterozygosity is 0.705(from 0.541 to 0.817).Thepolymorphic information of all the microsatellite markers are at a high or middle level,socan supply a credible information for this study.The microsatellite markers-phenotype correlative analysis uncovered that 9microsatellite markers had a significant or extreme significant impact on part of wooltraits.They are as following:the marker BMS0460,BM3501 and BL0004 have a significantimpact on Staple length (p<0.05),and the marker AE25 is extremesignificantly(p<0.01);the marker ILSTS22 has a significant impact on staple length at 18months (P<0.05);the markerOarcp43, MILVET09 have a extreme significant(p<0.01) andsignificant (P<0.05) impact on wool yield at 18 months;the marker RM150, ILSTS22,ILSTS0049 have a significant impact on wool yield at 30 months (P<0.05),and the markerMILVET09 is extreme significantly(p<0.01);the marker Oarcp43 have a significant impact on wool fibre diameter at 30 months (P<0.05) ,and the marker AE25 is extremesignificantly(p<0.01).A multiple comparison was made between genotypes and the markers which aresignificantly correlated with the phenotype trait.The results are as following:in the markerBMS0460,allele G has additive effect to staple length,and genotypes GG,DG, BB are thedominant genotype; in the marker Oarcp43,genotype BD is the dominant genotypes forwool yield at 18 months, genotype DD is the dominant genotypes for wool fibre diameterat 30 months; in the marker BM3501,genotypeⅡis the dominant genotypes for staplelength,and alleleⅠis the dominant allele;In the marker RM150,allele F has additive effectto wool yield at 30 months,and allele C is depletive, and genotype AF is the dominantgenotypes;in the marker BL0004, genotypes CE,BC and EE are the dominant genotypesfor staple length;in the marker ILSTS22,genotypes DD,EF and DE are the dominantgenotypes for staple length at 18 months,allele D is the dominant allele. Genotype FF isthe dominant genotype for wool yield at 30 months, allele F has additive effect to woolyield at 30 months,and allele E is depletive;in the marker MILVET09, genotypesBB,AC,BC are the dominant genotypes for wool yield at 18 months and wool yield at 30months;in the marker AE25, genotype EG is the dominant genotype for staple length,andgenotype EF is the dominant genotype for wool fibre diameter at 30 months;in the markerILSTS0049, AG is the dominant genotype for wool yield at 30 months.
