This paper mainly dealt with the study on the preliminary conservation of germplasm resources of endangered plant, Cercidiphyllum japonicum Sieb.et Zucc. For know the better medium and proceeding of culturing Cercidiphyllum japonicum Sieb.et Zucc, we use orthogonal experimental design and analysis of variance to establish the technique system of tissue culture. The affected factors of propagation, existing problems in this technique and the corresponding solutions are also described, which provide the theoretical base and technical measures for industrial producing of plantlet. We study the promotion of exogenesis hormones treatment and its concentration and soaking time in the green shoot cutting and the woody cutting, to provide the theoretical base for cuttage rooting.(1)The study of tissue culture technique and the better medium for various stages are as follows:(1)April is the best season to draw materials, in which the germination rate are 98.33 %, 81.67% and 95.00% by stem, laminae and petiole respectively, the contaminated rate only are 25.00 %, 18.33% and 21.67% by stem, laminae and petiole respectively.(2)During the trial: 7 minutes is good to treat the stem by HgCl2(0.1 %) and 5 minutes is good to treat the laminae and petiole.(3)Initial culture:Initial medium of stem: MS+BA 0.5 mg·L-1+ 2, 4-D 0.01 mg·L-1,in which the germination rate is 100%, the callus induction rate is 96.67%, and the shoot forming index is 1.9 .Initial medium of laminae: MS+BA 1.0 mg·L-1+NAA 0.01 mg·L-1+2, 4-D 0.1 mg·L-1.in which the callus induction rate is 78.33% ;Initial medium of petiole: MS+BA 0.5 mg·L-1+NAA 0.05 mg·L-1+2, 4-D 0.1 mg·L-1,in which the callus induction rate is 83.33%.(4)Multiplication culture: MS+BA 1.0 mg·L-1+2, 4-D 0.01 mg·L-1+NAA 0.1 mg·L-1,in which the multiplication rate is 4.2 and the multiplication of efficiency seedings is 3.9 .
