Pinellia ternate polysaccharide is the main component of Pinellia ternate, Pinellia ternate polysacchafide was found to be anti-tumor activity, has an activity of activating netlike inner-skin system and could restrain the coming forth as well as the proliferation of tumour. In order to probe into the mechanism of the antirtumor of the pinellia temate polysaccharide, In this paper, the anti-tumor activity of Pinellia temate polysacchafide is studied by the way of studying its structure, the way of evaluating its ability of scavenging the free radical in vivo and vivio and antitumor acitivity. The extraction of pinellia temate polysaccharide using the method of Ultrasonic and precipitated by the addition ethanol, the structure using GC and FITR, and the antioxidation using four system of antioxidation and antitumor by way of the model of mice, the resuls shown that, the antitumor activity of pinellia ternate polysaccharide has the osculation relation with their tructure of,β-D-Glucan, and has the antirumor by the way of scavenging the free radical in vivo and vivio.The main results of this paper are summarized as the following three aspects:(1)The crude Pinellia ternate polysaccharide PT-0 is extracted from Pinellia ternate by means of Ultrasonic and precipitated by the addition ethanol, and in a yield of 1.538%. Three kinds of polysaccharides are obtained from separating PT-0 by adding CTAB in a yield of 25.8%, 16.7%and 22.75% separately. The Phenol-sulfuric acid method is used to measure the total sugar contents of the above four kinds of polysaccharides, and the total sugar content are 83.37g/100g,95.04g/100g,97.63g/100g,90.26g/100g. PT-2 appears to be white spiky crystal and the others are all white powder. All the polysaccharides are dissolvable in water but not in organic solvent. Results of the UV-spectrophotometry show that none of the four polysaccharides contains impurities like nucleic acid and protein. The solubility of the polysaccharide by derived from the crude polysaccharide Pt-0 was accreted. The FT-IR spectrophotometry indicates that all of the polysaccharides are pyranose form, andβ-configuration of D- glucan, and have a structure ofβ- heteromorphism. Studies by GC show that PT-0 is mainly made up of xylose,mannose,fructose,Galactose,Glucose, that PT-1 is made up of xylose, Mannose, Glucose, that PT-2 is made up of xylose,fructose,Galactose,Glucose,that PT-3 is made up of cascaraose, xylose,Mannose, Galactos,Glucose.(2) The capacity of deoxidizing of Pinellia ternate polysaccharide by using the system of K3[Fe(CN)6] and TCA, the antioxidative of lipid by using the system of the Peroxidation of Polyunsaturated Fatty Acid from lipoprotein Induced by Iron., the activities of scavenging "OH radical by using the system of 1,10-Phenanthroline monohydrate and the activities of 02 radical of aforementioned polysaccharides by using he system of alkaline pyrogallol. Results show that all the polysaccharides have he capability of deoxidizing themselves, restrainting the lipid peroxidantion, and eliminating the 02-and "OH free radical. Moreover, the ability of antioxidation of PT-2 is enhanced with a increasing consistency, all kinds of Pinellia ternate polysaccharide have the activity of antioxidation, but the AP2 have the good relation between the quantity and the effect.(3)The model of mice is used to study the anti-tumor activity of PT-0 and PT-2. Results show that both of the polysaccharides can improve the immune ability of the body and that PT-O remarkably restrained the growth of the tumor and the effective Inhibition ratio is higher than that of PT-2. The contents of SOD, GSH-PX, CAT and MDA in liver, heart, brain and serum of the mouse are investigated. Results indicate that PT-2 can affect the SOD activity and has a target-effect. The improvement of activity of GSH-PX in liver and heart by PT-0 is a consequence of cooperative work of multi-polysaccharides, but in brain and serum this improvement is caused by the polysaccharides except PT-2. Meanwhile, PT-2 has a noticeablely influence on the activity of CAT in the heart of the mouse. The contents of MDA in liver and heart of the mouse can be depressed if the concentration of PT-0 and PT-2 reach a certain value and the effective concentration of PT-O is lower than that of PT-2, which indicates that PT-0 has a better ability of restraint. The effect of multi-polysaccharides may account for this phenomenon. PT-2 can essentially reduce the MDA in the brain of the mouse and specialtily reduce that in the serum of the mice.This study shown that, the antitumor activity of the Pinellia ternate polysaccharide osculated with their structure.
