Effect Of Shengjin Yu - Li Decoction On Immune Function In Patients With Primary Sjogren 's Syndrome Of Qi - Yin Deficiency Syndrome

Purpose： To research the immune function in Sj gren’s Syndromepatients who were diagnosed qi and Yin deficiency in TCM and treated bythe decoction of herbs producing body fluid.Method：Select42patients with primary Sjogren’s syndrome (PSS) anda diagnosis of qi and Yin deficiency in TCM in our hospital rheumatologyoutpatient or inpatient in the test,who are randomly divided intotreatment group and control group (n=21),and20cases as the healthycontrol group selected from healthy examination. The treatment group aretreated with the decoction of herbs producing body fluid, and the controlgroup are given routine clinical treatment of traditional Chinesemedicine extract radix paeoniae alba total glycosides capsules. Analyzethe peripheral blood T cell subsets by Using flow cytometry, and test theserum IL-17, IL-10level by the enzyme-linked immuneosorbentmethod.Analyze the T cell subsets, IL-17and IL10-level of patients withhealthy, then analyze the influence of T cell subsets, IL-17and IL–10in patients after treatment.Results：1. Before the treatment:the level of IL-17in the serum of patients was39.06±55.47ng/ml，significantly higher than that of the healthy controls10.04±2.71ng/ml (p=0.017).The level of IL-10in the serum of patients was21.97±6.96ng/ml，significantly lower than that of the healthy controls39.86±10.13ng/ml (p=p=0.000)．The percent of CD3+T cell in the bloodof patients was68.71％±8.12％，higher than that of the healthy controls 66.10%±4.18％，but without statistical difference(p=0.246). The percentof CD4+T cell in the blood of patients was36.90%±8.31%，significantlylower than that of the healthy controls42.90%±3.32%(p=0.037)．Thepercent of CD8+T cell in the blood of patients was28.67%±10.66%，significantly higher than that of the healthy controls18.40%±2.55%(p=0.000)．The level of CD4/CD8in the blood of patients was1.61±0.72，significantly lower than that of the healthy controls2.4±0.42(p=0.003)．2. By comparing the two groups of test indexes before and after treatment:IL-10in the treatment group after treatment was significantly increasedcompared with before treatment（p＜0.05）,and in the control group aftertreatment there was no significant difference than before treatment（p＞0.05） but it had present a rise trend. There was no significantdifference in IL–17of the two groups before and after treatment. Butbefore and after treatment the treatment group was38.94±66.49vs.29.49±43.74(pg/ml) and control group was39.17±45.57vs.31.92±30.78(pg/ml), both presenting a downward trend. There was no significantdifference in T cell subsets of the two groups before and after treatment.But before and after treatment the CD8in treatment group was30.2±12.18VS28.7±9.43（%） and in control group was28.8±8.40VS27.8±7.30（%）, both presenting a downward trend. The CD4/CD8in treatment groupwas1.41±0.59VS1.49±0.58（%） and in control group was1.29±0.39VS1.36±0.36（%）, both presenting a rise trend. The IgG in the two groupsafter treatment were significantly increased （p＜0.01）,and IgM and IgAwere decreased (p <0.05). The anti SSA and anti SSB antibody positiverate decreased after treatment, but there was no significant difference(p＞0.05).3. The treatment group total effective rate was76%and control group total effective rate was71%by treating with the decoction of herbs producingbody fluid.Conclusion：1. The number of IL-17in the serum of patients was significantly higherthan normal, and the number of serum IL-10of patients was significantlylower than normal, which suggested the qi and Yin deficiency syndrome inpatients with PSS has immune dysfunction and chronic inflammation.2. By treating with the decoction of herbs producing body fluid, therewas no obvious statistical significance in T cell subsets, but CD8+Tcells was on the decline and CD4/CD8was on the rise, which suggestedit had regulation of T cell subsets.3. After the treatment with the decoction of herbs producing body fluid,IL-10significantly increased and IL-17was on the decline, reminding thatthe methods have the two-way function of the regulation effects oninterleukin.4. The immune globulin in patients, SSA antibody and SSB antibody positiveexpression were reduced by using with the decoction of herbs producingbody fluid to treat PSS in qi and Yin deficiency syndrome, which can adjustthe immune function.5. There was effective treatment in PSS with the decoction of herbsproducing body fluid. It can improve the clinical symptoms and regulateimmune function to achieve the efficacy of treatment.
