Fan flower is a group of characteristic flowers in Australia, known with their fan-shape flowers and belonging to Goodeniaceae, Scaevola. Scaevola crassifolia Labill is one of the species in this genus. This paper researched the tissue culture and cutting propagation of S. crassifolia, in order to provide technique supports for its landscaping application. Main results are as following:(1) Stems segments with axillary buds were chosen as explants in different season and were sterilized with two-step sterilizing method. Sterilizing experiment showed that it was better to pick explants in early summer than in winter. Best result were obtained by sterilizing explants picked in April to May with 1% NaClO for 20 minutes, which percentages of contamination, death and shoots germination were 32.41%,17.57%and 50.02% respectively.(2) Aseptic shoots were obtained via axillary buds multiplication. The best medium for axillary buds multiplication was MS without hormone with a bud's germination rate of 100%.(3) The best medium for directly shoots regeneration from aseptic leaves and petioles were both MS+0.2mg/L BA, and the effective shoots induction rate were 50.01% and 25.02% respectively.(4) The four-factor and three-level orthogonal experiment for callus induction showed that the best callus induction could be obtained by culturing petiole explants in the medium of MS+0.2mg/L BA+0.5mg/L 2,4-D in dark for 30 days, and the callus induction rate reached 86.04%.(5) The optimum medium for adventitious shoots induction from callus was MS+0.2mg/L BA, and the effective induction rate was 79.53%.(6) The best medium for shoots proliferation was MS+0.4 mg/L BA.(7) Vigorous shoots could be obtained by culturing the shoots in MS medium with 30g/L sucrose.(8) The four-factor and three-level orthogonal experiment for rooting showed the best rooting medium was 1/2 MS+0.5mg/L IBA+0.1mg/L NAA+30g/L sucrose, and the rooting rate was 100%, the average roots length was 15.7cm.(9) Turf soil:vermiculite:ruby mica=1:1:1 was the best transplanting medium, and the survival rate was 47.37%, the transplant seedlings grew well.(10) NAA was optimum for cutting propagation. The rooting rate, roots number and roots length reached the highest when NAA concentration was 1500mg/L.(11) Cutting plantlets transplanted in garden soil grew faster and better with higher survival rate than in other transplanting medium.
