The Plant Regeneration And Rapid Propagation Of Orostachys Fimbriatus

The orostachys fimbriatus (Orostachys fimbriatus) is belong to the crassulaceae tile pine plant, and grows on the roof tile seam or on the rock. The organic acid compounds of the plant include cardiac glycosides, flavonoids, oxalic acid, gallic acid and so on, commonly it is used to treat illness, for example cervical erosion, hemorrhoids, mouth ulcers.In this study, tissue culture engineering for wild orostachys fimbriatus as explants is been researched. The regeneration system has been modified by studying the effect factors on regeneration such as the different drawn time, the processing method, the disinfection time, the proportion of hormone etc. The main results are as follows:1. The material selects for orostachys fimbriatus aseptic system should be early in May, drawing materials heel after processing, cutting the new germination of the stem segments as the explants. The disinfectant is 0.1% mercuric chloride for 10min, the rate of pollution is below 5% and the survival percentage is above 92%. Using the MS+NAA0.1mg/L+6-BA0.5mg/L for start culture medium, the germinative rate is 99.78%, and the phenomena of the callus and vitrification do not appear.2. Cutting the stem tips and the stem section in initial medium to culture with MS+6-BA1.0mg/L+IBA0.2mg/L+sucrose20g medium. The stem tips reproducibility reaches 1386%, the bud seedling is not only vigorous, but also grows few short roots; the stem section reproducibility is to 1169%, the average value of rooting is 4, and the proliferous buds grow healthy. MS+6-BA1.5mg/L+IBA0.1mg/L+ sucrose 30g is the optimum medium for the leaves, which grows some small adventitious buds, the reproducibility reaches 971%.3. Selecting vigorous proliferous buds and cutting a single stem segments to culture with 1/2MS + IBA0.3mg / L + NAA0.05 ~ 0.1mg / L medium. The average value of rooting is from 11 to 13 and the plant grows vigorous.4. Selecting the orostachys fimbriatus with complete root system to place outdoors and re-place the bottle cap after opening the bottle and ventilation. Re-opening the bottle to train the seedling for 5~9d after sealing to exercise the seedling for 3d, the survival rate of seedling was significantly higher and the lowest survival rate is 95.5%. The seedlings grow in the medium with edible fungus polishings and vermiculite = 1:1 ~ 1.5 after training, the survival rate of seedling is 99.3%.
