To evaluate the effects of Chinese herbal medicine extracts on proliferation of normal mouse mammary epithelial cells in primary culture, the mammary tissue clumps from KM mouse in lactation period at adulthood were obtained and cultured to harvest epithelial cells. The morphological identification of normal mouse mammary epithelial cells in primary culture was determined under inverted microscope. The cell population doubling time and the cells growth curve were observed and investigated. The lactation function of the cells was detected by SDS-PAGE. Folium Hibisci Mutabilis, Herba Violae, Rhizome Paris, Herba Lycop, Herba Scutellaria Barbata and Chinese medicine compound prescription were extracted to add to cell cultural medium to study their effects on multiplication of mouse mammary epithelial cells.The results were as follows:In this study, primary culture of mice mammary gland cells could be derived by outgrowth of migrating cells from fragment of tissue, which were treated by 0.25% trypsin in Hanks for 35 min at 37℃. Fibroblast and epithelial cells could be pured according to their different sensibility to trypsin. Complex cultures were treated by 0.25% trypsin in Hanks for 57 min at 37℃, the dispersed cells were mainly fibroblasts, then the rest was treated by 0.25% trypsin in Hanks for 58 min at 37℃, the majority cells harvested were mammary epithelial cells. The pured mammary epithelial cell line could be obtained by 23 Passages.The result indicated that the Chinese herbal medicine compound preparation could promote the multiplication of mouse mammary epithelial cells at the consistency of 1.6 mg·mL-10.2 g·mL-1 (P<0.05), especially at the consistency of 0.2 g·mL-1, as the consistency reduced, the effect of promotion become unconspicuous (P>0.05).Rhizome Paris could inhibit the multiplication of mouse mammary epithelial cells at a high consistency (0.2 g·mL-1, p<0.05), but as the consistency lowed down (40 mg·mL-1, 320μg·mL-1), the promoting multiplication effect appeared (p<0.05), without any linear relationship. Folium Hibisci Mutabilis appeared two consistency (0.2 g·mL-1, 8 mg·mL-1) of inhibit the multiplication of mouse mammary epithelial cells and two consistency of promoting it (1.6 mg·mL-1, 40 mg·mL-1), both are significantly different from control group (P<0.05). Herba Lycop could promote the multiplication of mouse mammary epithelial cells under the consistency of 40 mg·mL-1 (p<0.05), and inhibit the multiplication at the consistency of 0.2 g·mL-1(p<0.05). Herba Scutellaria Barbata could inhibit the multiplication of mouse mammary epithelial cells at a high consistency (0.2 g·mL-1, p<0.05), but as the consistency lowed down (1.6 mg·mL-18 mg·mL-1), the promoting multiplication effect appeared (p<0.05), especially at the consistency of 1.6 mg·mL-1. Herba Violae did not appear the effect of inhibition, and the promotion peak value was closed to 40 mg·mL-1 (p<0.05), and the multiplication-promoting effect become no sighificant as the consistency lowed down.The results showed that Chinese herbal medicine extracts and compound preparation could promote the multiplication of mouse mammary epithelial cells at the right level, but if the dose was too high or medication time was excess, the multiplication of mouse mammary epithelial cells was inhibited.
