The effect of hormone on the callus induction and differentiation of cotyledon,leaves and stem of Ulmus macrocarpa var. mongolica was studied. And an efficiency regeneration System of the plant was established. Through measuring SOD activity and daily growth rate of callus weight in order to pick the best medium and the proportion of hormone concentration, we finally assured the best Somatic embryogenesis system. The results were followed:1. The inductivity of the MS medium was obviously higher than the LP,DCR medium's in the process of callus induction.2. By comparative experiment among three differentiation explants such as cotyledon leaves and stem, we found the best differentiation explants of callus of Ulmus macrocarpa var. mongolica was cotyledon.3. The best proportion of medium of callus induction of Ulmus macrocarpa var. mongolica are: MS+2,4-D 1.5 mg·L-1+6-BA 0.2 mg·L-1.4. The proliferated callus leaded to raise SOD activity, speeded up metabolism and increased growth vigor of callus. Therefore, we can identify growth rate of callus of Ulmus macrocarpa var. mongolica by the index.5. The best proportion of medium of callus differentiation of Ulmus macrocarpa var. mongolica was: MS+IAA 5.0 mg·L-1+6-BA1.0 mg·L-1.6. The best proportion of medium of rooting of Ulmus macrocarpa var. mongolica are: 1/2MS + IBA 0.4 mg·L-1.7. The best regeneration hardened matrix of Ulmus macrocarpa var. mongolica are: vermiculite + perlite + peat soil(1:1:1).
