The paper takes the stem segment, the tissue-cultured leaf and the tissue-cultured leaf rachis as explants, studies the establishment of tissue culture technique regeneration system for Sorbus aucuparia。The main contents and results are listed as follows:(1) The disfinfections of explants are different obviously。The best disfinfection of the spring stem segment is the Saturated calcium hypochlorite, 15min, but the summer stem segment needs 30min and the disfinfections is poorer。The disfinfections of the summer stem segment with 0.1% HgCl is better than the disfinfections with the Saturated calcium hypochlorite, the best time is 8min。Generally speaking, the disfinfections of the spring stem segment is better than the summer stem segment。(2) The optimum medium for the stem segment germinating is MS＋6－BA2.0 mg/L＋0.1 NAA mg/L。The germination rate of the spring stem segment and the summer stem segment is 89.47% and 64.00%。(3) The optimum medium for the multiplication of the regenerating plantlets is 1/2MS＋6－BA1.0mg/L＋NAA0.2mg/L。(4) The optimum medium for leaf callus inducing is MS＋6－BA2.0mg/L＋NAA0.1mg/L or MS＋6－BA1.0mg/L＋IBA0.05mg/L, for the leaf rachis callus inducing is MS＋6－BA2.0mg/L＋NAA0.05～0.1mg/L or MS＋6－BA2.0mg/L＋IBA0.05mg/L。The callus in these medium grows well and multiplicats quickly, but it doesn't induce the bud; The culture under dark is no influence for the leaf and the leaf rachis callus inducing。(5) The optimum medium for the duplicate leaf regenerating bud is MS＋BA2.0mg/L＋KT1.5mg/L＋NAA0.05mg/L, for the leaf rachis regenerating bud is MS＋BA1.0mg/L＋KT 1.5mg/L＋NAA0.05mg/L。The regenerating buds in these medium grows vigorous and healthy, and can root after muttiplicating。The culture under dark is advantageous in the duplicate leaf and the leaf rachis regenera-t ing bud, and can enhance the regenerating bud rate and the NO.of buds on per duplicate leaf and leaf rachis。The best time is three weeks。(6) The optimum medium for rooting is 1/2MS＋IBA0.01mg/L or 1/2MS＋2, 4-D 0.03mg/L。The rooting rate is the highest and the regenerating plantlets grows healthy。(7) The survival rate of the plant is 88.90% after domesticating and transplanting。...
