Monosomic Additions Of Beet M14 Tissue Culture

Using the fresh leaves,leafstalks of plants and leaves,eustipes of aseptic seedlings of Sugar Beet M14 as the explants,which were inoculated on MS medium supplemented with different hormone and nourishing substances,such as 6-BA,NAA,IBA etc.,we set a technical tissue culture rapid propagation system for Sugar Beet M14,studied the main factors which influenced the tissue culture and regeneration,and stressed the best medium and proceeding during the process.The main contents and results are listed as follows:1,Using the leaves,leafstalks of plants and leaves,eustipes of aseptic seedlings of Sugar Beet M14 as different explants,after filtration and original culture,it is found that regeneration ability of eustipes of aseptic seedlings is the best.The optimizing medium for inducing adventitious buds is MS+6-BA 2.0mg/L+NAA 0.1 mg/L,and the inductivity is 93.9%.These adventitious buds grow well,and they can root after transferred to medium of rooting and can become integrated plantlets.2,The optimizing medium for leaves explants inducing callus is MS+6-BA 2.0mg/L+NAA 0.5mg/L,and the inductivity is 75.3%.The optimizing medium for leafstalk explants inducing callus is MS+6-BA 2.0mg/L+NAA 0.5mg/L,and the inductivity is 90.9%.Calluses are all incompact in a light chartreuse color.These calluses did not continue to produce adventitious buds after induction.3,The ideal bourgeon condition for seeds in an aseptic environment is:darkness, MS+NAA 0.5mg/L,and the inductivity is 93.2%.4,The best medium for regenerative plantlets multiplication is MS+6-BA 1.0mg/L+NAA 0.05mg/L.With this concentration,regenerative plantlets grow healthily,propagate rapidly,and the leaves are green.Proliferation times achieve to 3.78. The research of other culture condition displayed that there was no significant difference for the 4 basic culture mediums.The optimal length of illumination,the optimal intensity of illumination and the optimal concentration of sucrose are 14h/d, 20001x and 20g/L respectively.5,M14 is a not-easy-root plant.The best medium for rooting is 1/2MS+IBA 3mg/L,and the best inductivity is only 50%.Roots are thin and weak as well.
