The wheat took the world first big grain crops, have the obvious heterosis.Use its heterosis effectively, is an important way to enhance the wheat output. At present the wheat heterosis use mainly has three ways. With a long time developed, all has made the progress, but still has many problems in the practical application. Non-1BL/1RS male sterile line of NIAN type in wheat completely overcome the shortcomings of the past many types, in particular greatly enhanced its line of easy recovery and restoration. This study will address these merits, to push the non-1BL/1RS male sterile line of NIAN type forward as soon as possible, particularly the application for its use. To provide the theoretical information and technical support.In this paper, the methods of single-minded hybridity, advanced SDS-PAGE analysis and microscope were applied to transduce gene rfv1 that is the male sterile gene from T.machavar. subletschchumicum and T.spelta var.duhamelianum , into the targeted female parents. The important results obtained are as follows:1 Improvement of identification ways.Based on the different chromosomes, we have the different ways to identify the hybrids.Meanwhile, the author have done a lot of pre-experiments to improve the identification of methods, to give the smooth progress of experiment, make the results reliable and accurate. In the identification process, the link between all ways connected and complement each other. Ensure that the results of the identification of the true and accurate.2 In the respect of rfv1 into1BL/1RS CMS. Based on the donor material for non-1BL/1RS chromosome characteristic, The experiment take the 1BL/1RS male-sterile line Fp1 as the acceptor. The difference in chromosome causes the hybrid and the backcross descendant available to microscopic exam appraisal. Use the SDS-PAGE electrophoretic analysis, confirms the rfv1 gene 's appear.From this may, when we use the non-1BL/1RS female parent Fp2 as the acceptor. SDS-PAGE electrophoretic analysises provid accurate and the simple appraisal method.3 In the respect of rfv1 into non-1BL/1RS CMS. As for the body, both donor and receptor has non-1BL/1RS chromosome, Therefore, the use of microscope can not observed their differences. They also have the same band in SDS-PAGE electrophoresis analysis. So we re-select 'SP4' as the donor. Using SDS-PAGE analysis for hybrid and backcross progeny. Meanwhile, we use the traditional method of mating to test the SDS-PAGE results.4 The study has the great significance.We can obtained a new non-1BL/1RS male sterile line with 'Xinong Fp1' agricultural characters. It promote wheat heterosis for the use of new technology system. From this mew techology, the great potential production of hybrid wheat can be applied practically and also promoting the development of the male sterile wheat lines of NIAN type in wheat.
