Northeast forest frog (Rana dybowskii) taked an important economic place in the northeast of China, and it was one of the most important medical animals in Heilongjiang Province. As an organ which contact directly with habitat, skin of the frog had been formatted gradulatly a natural line to defense microbes during evolutionary. Recent research of Northeast forest frog focused on nutrient content and the structure and function of activitai peptide of skin, while the research in the state of stress in morphology of skin and cutaneous glands had been seldom reported. The purpose of this article was to comparatively observe the change of morphology and the distributing of cutaneous glands in the northeast forest frog in different environmental stress using the technology of histology and immunocytochemistry. And this would offer basal information of the relation between habitat environment and its function in morphology structure and the distributing of cutaneous glands in the frog.The northeast forest frog which had been used were from Tieli of Heilongjiang province; The method of Histology and Immunocytochemistry were been used to observe the adaptive change of normal state, low temperature stress, electrical stimulation and injected noradrenalin in lymphocyte.The results showed that it was differe.nt in the thick of the skin, layer of cell , the distributing of the glands and the density of chromatophores cell among different part. The total thickness of dorsal skin was thicker than ventral skin. The skin was formed by epidermis and dermis. There were chromatophores cell between them and the color in ventral skin were bigger than that in dorsal in volum. Granular distributed more in the ventral skin and granular glands was mostly existed in the dorsal skin, biggest of which was between the two lines of dorsal body in volumn. Mucus glands was distributed widely all over of the skin, more in ventral skin.The sizes of granular glands were all shorter in low temperature stress than that in normal states. Statistics showed that significant difference was only in C. The quantity rate of granular glands were lower than normal strate, which distinct in the back. The length of granular glands were minish with the time pass by after electrical stimulation, which were lowest at 90min and 120min.And The rate of granular glands gradually reduced to the lowest after 1 day out. And then accretion rapidly. It had significant difference in the length of granular glands of skin in Northeast forest frog after injecting horadrenalin(P<0.01).The results showed that bombesin were mainly distributed in the plasm of mucus gland.The results of the method of SABC indicated that bomesin of the northeast forest frog express weal in the natural or injecting noradrenalin, but express strong in the low temperature and electrical stimulate for 0min,30min, expressing stronger for 60min, expressing weak for 120min,1d,2d,4d, expressing weaker for 6days.
