Establishment Of Regeneration System Cultured In Vitro Of Agapanthus Praeco Ssp.orientalis 'Big Blue'

Using the fresh leaves, roots and pedicels etc. as the explants, which were inoculated onMS medium supplemented with different hormone and nourishing substance, such as 2,4-D, 6-BA, NAA, IBA etc., we set a technical tissue culture rapid propagation system for Agapanthuspraeco ssp.orientalis'Big Blue'(SVH) for short, studied the main factors of influencing thetissue culture and regeneration, and stressed the best medium and proceeding during theprocess. The main contents and results are listed as follows:(1) Using leaves, roots and pedicels as different explants, after filtration and originalculture, the results indicates regeneration ability of young embryo is the best. The optimizingmedium for inducing callus of young embryo is MS+6-BA1.0mg/l+2,4-D1.0mg/l+KT0.05mg/l, inductivity is 78.3%. The optimizing medium for inducing adventitious buds is MS+6-BA2.0mg/l+IBA0.1mg/l, inductivity is 86.4%.These adventitious buds grow well, they canroot after transferred to medium of rooting and can become integrated plantlets.(2) The optimizing medium for pedicels explants inducing adventitious buds is MS+6-BA2.0mg/l+IBA0.1mg/l, inductivity is 59.6%.(3) The optimizing medium for explants of roots is MS+6-BA0.2mg/l+PIC1.0mg/l,inductivity is 59.6%. Callus is incompact in a light chartreuse color. These callus doesn'tcontinue to produce adventitious buds after induction.(4) The ideal bourgeon condition for seeds in an aseptic environment is: under darkness,MS+NAA0.5mg/l, inductivity is 86.2%.(5) The best medium for regenerative plantlets multiplication is MS+6-BA1.0mg/l+NAA0.05mg/l。With this concentration, regenerative plantlets grow healthily, propagate rapidly,leaves are green, proliferation times achieve to 3.56.(6) The best medium for rooting is 1/2MS+IBA0.1mg/l, roots are healthy, thickset andgoodliness, the regrowth grows well. Agapanthus praeco ssp.orientalis 'Big Blue' can rooton medium without any hormone, it's a easy-root plant.(7) When tissue culture plant from adventitious buds is 2cm-high, the best health youngplants are chosen to be toughened. Firstly, we open seal membranes on triangle bottles for 3-5days, cultivate young plants for 5-7 days in the water. Then, the plants are cultivated invermiculite. Pay attention to overshadow and irrigate on time, The survival rate reaches 93.3%.
