Study On Meat Performance, Correlation Between A-FABP,EX-FABP Gene And IMF Contents Of Kirin Chicken(Frizzle Chicken)

The research took Frizzle Chicken as the research material, determined and compared growth performance, slaughter performance, blood physiological and biochemical indexes of the male and the female at 16-week old; then used soxhlet apparatus to determine intramuscular fat content of Frizzle Chicken, and with A-FABP and EX-FABP gene as candidate gene, using direct sequencing method and PCR-SSCP method to detect the candidate gene single nucleotide polymorphisms, explore the intramuscular fat content relevance to the different genotype combinations of the Frizzle Chicken; finally, used RT-PCR to analyse A-FABP and EX-FABP gene in Frizzle Chicken 12, 14, 16 weeks m RNA expression in the organization and correlation with intramuscular fat. The research aimed to provide theoretical parameters and lay the foundation of Frizzle Chicken’development and utilization in tropical heat areas. It also provided scientific basis and reference for the reserach of poultry and other animals’ intramuscular fat. The results were as follows:1. Meat performance(1) Growth Performance: At the end of 16 weeks, the male bird average weight was 1717.89 g, and the female bird average weight was 1475.97 g.In the fifth week, the male absolute growth rate reached a maximum of 29.27 g / day, the female also reached a maximum of 24.12 g / day. The relative growth rate of the male and the female were both gradually decreasing with the breeding time. The male was less than female in feed conversion ratio(P>0.05). The feed conversion ratio of the male and the female(except the 14 weeks old) increased with time increasing.(2) Blood Physiological and Biochemical Indexes: It had significant(P<0.05) or extremely significant(P<0.01) between the male and the female in MCHC, RBC, HGB, HCT and MCH. The male of CK level was up to 5242.57 U / L, the female was up to 5524.02 U /L.(3) Slaughter performance: The coefficient variations of the 4 body size in the male and the female were both within 10%. In the slaughter performance,only eviscerated ratio and four fat traits(including abdominal fat weight, abdominal fat ratio,intramuscular fat wide and sebum thick)showed that the female weresignificantly(P<0.01) higher than the male.2. SNP: It both existed one polymorphic site in A-FABP gene exon 3 1763A> G sites, EX-FABP gene exon 2 3086A> G site and exon 4 of the 4271G> A sites. Correlation analysis with breast IMF content and leg IMF content, results showed that BB type,CC type and FF type respectively were advantage genotypes of breast IMF content;AA type, BB type and EF type respectively were advantage genotypes of leg IMF content. In breast IMF content, the combination genotypes of BBCC and BBCCEF were predominant genotypes; ABDD, DDEF and ABDDEF were disadvantages genotypes. In leg IMF content, the combination genotypes of AACC and AACCEF were predominant genotypes; ABFF and ABCDFF were disadvantages genotypes.3. RT-PCR: The amplification efficiency E of the target genes A-FABP, EX-FABP and the reference gene β-Actin were 99.34%, 97.93% and 94.26%. There were significant difference(P<0.01) between 12 th week, 14 th week and 16 th week in the male A-FABP gene relative expression levels in each tissue. The female A-FABP gene relative expression of breast muscle in 12 th week, 14 th week compared with the 16 th week existed a significant difference(P<0.01),and the relative expression of 12 th week had a significant difference(P<0.05)with 14 th week;In the leg, heart and liver, the relative expression had significant differences(P<0.01)between 12 th week, 14 th week and 16 th week. In the male EX-FABP gene relative expression levels of breast muscle, the 12 th week and 16 th week compared with 14 th week existed a significant difference(P<0.01), the expression of 12 th week and 16 th week had no significant difference(P>0.05); In leg muscle, the 12 th week and 14 th week compared with 16 th week existed a significant difference(P<0.01), but 12 th week and 14 th week had no significant difference(P>0.05);In the heart, the 12 th week and 14 th week compared with 16 th week existed a significant difference(P<0.01), and the 12 th week and 14 th week had a significant difference(P<0.05); In the liver, there were significant difference(P<0.01) between 12 th week, 14 th week and 16 th week. In the female EX-FABP gene relative expression levels, there were significant difference(P<0.01)between 12 th week, 14 th week and 16 th week of breast muscle and hear; the 12 th week and 14 th week compared with 16 th week existed a significant difference(P<0.01),but the 12 th week and 14 th week had no significant difference(P>0.05).
