Quarantine Research Of Nematodes In Imported Wooden Package In Ningbo Port

Many pests can be carried by imported wooden packages during transportation, and nematodes, especially the pine wood nematode, are among our main concerns. This research is mainly on the nematode quarantine in imported wooden packages. There are three parts in this thesis: firstly, morphological study of Bursaphelenchus species in the imported wooden packages in Ningbo port, China; secondly, identification of Bursaphelenchus fraudulentus in Belgium oak tree based on both morphological characters and molecular data; last, analysis of quarantine on nematodes in the imported wooden packages in Ningbo port, China.Part I: Morphological study of Bursaphelenchus species in the imported wooden packages in Ningbo port, China. From January 2003 to December 2006, about 11768 batches of wooden packages had been inspected and identified for nematodes, especially for Bursaphelenchus species. Among them, 464 batches were identified to be infested by the species of Bursaphelenchus. According to their morphological and molecular characters, 14 species of Bursaphelenchus were identified and a key to these species is provided.Part II: Identification of Bursaphelenchus fraudulentus in Belgium oak tree. Based on observation of morphological characters and measurements, and by the method of PCR-RFLP, nematodes detected in the oak wood from Belgium were identified as Bursaphelenchus fraudulentus, which was the first report of this species in Ningbo port, China.Part III: Analysis of quarantine on nematodes in the imported wooden packages in Ningbo port, China in recent years. Based on large amounts of wooden packages inspection combined with the duty of Plant Quarantine Lab of Ningbo Port, Bursaphelenchus species related with wooden packages and related countries were analyzed. So this research has laid a solid base for the further Bursaphelenchus species identification in the imported wooden packages in Ningbo port, China.
