Dairy cattle mastiffs is a disease with high incidence rate,which could lead to great loss.The MIC(minimum inhibitory concentrations) of Chinese herbal medicines such as Phellodendron Amurense,Herba Taraxaci,Flos Lonicerae,Fructus Forsythiae,Tokyo Violet Herb,Herba Houttuyniae,Flos chrysanthemi indici were detected against the main pathogenic bacteria causing dairy cattle mastitis in vitro.Complex prescriptions were done the same as the former.Some medicines,such as Phellodendron Amurense,Herba Taraxaci,Tokyo Violet Herb,which had better antibacterial action than the others,were selected by the effect of MIC.Therefore,the three Chinese traditional medicines which had better antibacterial activities were chosen to be combined with Lumbricus,Common burreed tuber,Zedoary rhizome,Zanthoxylum bungeanum and to make the compound medicine Ruxiankang sticking plaster according to the Chinese veterinary medicine theories.The quality standard of the praeparatum,the efficiencies of alleviating pain, anti-inflammatory,relieving fever and improving microcirculation,the permeability to rat skin,the security and the clinic curative effect were studied.The results showed as follow.In the study of the quality standard,TLC was used to identify the compound medicine Ruxiankang sticking plaster.TLC results showed that the separation of the stains was apparent and the negative contrast was not disturbed.HPLC was used to determine the content of berberine hydrochloride,and the condition of chromatographic analysis of berberine hydrochioride was as follow:C18 column(250mm×4.6mm,5μm),the mobile phase consisted of acetonitrile-0.1% phosphoric acid-triethylamine(50:50:0.75) at a flow-rate of 1.0 mL·min-1,the ultraviolet detector was set at 265nm,the column temperature was 30℃.The results showed that berberine hydrochloride owned a good linear relationship at the range of 2.5~40μg·mL-1.The average recovery was 97.20%(n=6) and the RSD was 1.18%.All of them proved that the method used was specific.So the best method was gained which was the basis of exploring the quality control of berberine hydrochloride of compound medicine Ruxiankang sticking plaster.In the study of pharmacodynamics,the results of anti-inflammatory experiment showed that the compound medicine Ruxiankang sticking plaster with high and medial dosage could effectively inhibit auricle swelling in mice caused by xylene,swelling inhibitory rate was 59.6%,48.1% respectively and,compared with the model control group,the difference was significant(p<0.01). The compound medicine Ruxiankang sticking plaster with high and medial dosage could effectively reduce the formation of agar-induced chronic granulomatous inflammation in mice, swelling inhibitory rate was 31.73%,24.84%respectively and,compared with the model control group,the difference was significant(p<0.05).The results of alleviating pain experiment indicated that the hot plate-induced pain threshold was elevated in high,medial and low dosage compound medicine Ruxiankang sticking plaster groups,the pain threshold elevating rate was 111.23%, 92.47%,82.88%respectively and,compared with the model control group,the difference was significant(p<0.01).The results of antipyretic effect experiment showed that the compound medicine Ruxiankang sticking plaster with high,medial and low dosage had obvious effect on pyretic model evoked by sc turpentine oil in rat.The "blood-stasis" rarebit was prepared by adrenaline and ice water.Compared with the model control group,the whole blood viscosity, plasma viscosity,erythrocyte specific votume,erythrocyte aggregation index and erythrocyte transmutation index in Ruxiankang sticking plaster were significantly degraded(p<0.01,p<0.05). The results of skin-penetrating experiment showed that,with the time prolonging,the amount of berberine hydrochloride releasing and penetrating from the patch was increasing.The the rate of berberine hydrochloride penetration amounted to 39.64%within 48 hours.It suggested that skin-penetrating administration was available and could maintain invariable concentration in blood.In the aspect of toxicology,the transdermic LD50 of compound medicine Ruxiankang sticking plaster in rat was more than 45mg·kg-1.The results of skin hypersensitive experiments on cavy showed that the anaphylaxis rate of compound medicine Ruxiankang sticking plaster was 0%.The skin irritate test showed that,except the rabbits whose skin had been damaged showed slight suscitation,all the rabbits had no stimulus response.It showed that compound medicine Ruxiankang sticking plaster could be given through skin administrition.The cure rate of compound medicine Ruxiankang sticking plaster was 95.21%for clinical and 95.73%for subclinical dairy cattle mastitis,which is higher than the positive control medicine Ruyi-Jinghuang paste.And compound medicine Ruxiankang sticking plaster could increase the milk quality fast and effective indicating that compound medicine Ruxiankang sticking plaster possessed better efficacy for clinical and subclinical mastitis.In concludes,the compound preparation had obvious pharmacodynamic effects of antiinflammation, alleviating pain,antipyretic and improving microcirculation,and had a good penetration effect.The skin administer was safe.And it had better efficacy for clinical and subclinical mastitis.This study not only provided bases for the clinic application of the compound Chinese traditional medicine but also theoretical bases for developing effective medicine to treat dairy cow mastitis.
