| This experiment selected Dandelion, Honeysuckle Flower, Weeping Forsythia Capsule, Tokyo Violet Herb, Baical Skullcap Root, Isatis Root, Cape Jasmine Fruit seven herbs, in accordance with L8 (27) orthogonal table, determined the extent of inhibition of Herbs on each of Staphylococcus aureus, Escherichia coli, Streptococcus mastitidis using test-tube double dilution method, Finally we obtained the heat-clearing and detoxicating complex prescription of ultimate treatment of mastitis. Then used the xylene to send the mouse auricle swelling non-specificity inflammation model, we had studied the traditional Chinese medicine anti-inflammation function, and had determined the best traditional Chinese medicine composition on the basis of the heat-clearing and detoxicating complex prescription. The technologied process and the stability of traditional Chinese medicines ointment were studied, on the basis of traditional Chinese medicines pharmacy. The best enhancing penetration concentraction of azone in traditional Chinese medicines ointment were defined. The safety and clinical therapeutic action of traditional Chinese medicines ointment were studied.The heat-clearing and detoxicating complex prescription which obtained by in vitro antibiotic activity test result was: Baical Skullcap Root, Weeping Forsythia Capsule and Dandelion. The Baical Skullcap Root could enhance the complex prescription curative effect obviously, the dandelion played the inhibitory action in the complex prescription to the Escherichia coli and the Staphylococcus aureus, but did not have an effect to the Streptococcus mastitidis, the Weeping Forsythia Capsule played the inhibitory action in complex prescription to the Escherichia coli and Streptococcus mastitidis, but did not have an effect to the Staphylococcus aureus. The inhibitory effect of the ultimate heat-clearing and detoxicating complex prescription to Escherichia coli, Staphylococcus aureus, Streptococcus mastitidis was 1:64,1:32 and 1:32 respectively.The anti- inflammation test result indicated that the best complex prescription was composed of the heat-clearing and detoxicating complex prescription and the frankincense. The significant value of the heat-clearing and detoxicating complex prescription was 0.064 (0.05 0.10) respectively,The amounts of Azone in traditional chinese medicines ointment was determined 4%. The traditional chinese medicines ointment didn't appear any phenomena of breaking emulsion, mildewy and the change of colour, luster, homogeneity by cold-resistant, heat-resistant, centrifugal, accelerating experiment. The dried orange peel and the frankincense factor had non-statistics significance. showed that the heat-clearing and detoxicating complex prescription have certain influence to the change of tests index, the influence of other two factors to test index was not remarkable. And the influence order was in turn: the heat-clearing and detoxicating complex prescription > frankincense > dried orange peel.The results of irritative and anaphylaxis experiment showed that traditional Chinese medicines ointment were non- irritation or weaker anaphylaxis substance. When the traditional chinese medicines ointment was applicated externally, the intact or damaged skin of rabbit didn't appear the phenomena of erythema and oedema, and the score of skin irritative action were smaller than 0.5. The cavies in traditional Chinese medicines ointment group didn't appear the anaphylaxis symptom of erythema or oedema, and the anaphylaxis rate of traditional Chinese medicines ointment was 0%.The results of Clinical Pharmacodynamics experiment showed that the ointment had good therapeutic action on clinical mastitis of dairy cow curing and effective percent.The studies showed that traditional Chinese medicines ointment was a good skin permeating preparation, which was non-toxic, non- irritating, non-anaphylaxis, and had good curative effects on clinical mastitis of dairy cow. Its applicative scheme was that mammary gland were daubed 10~15g per times, and 2 times per day. Its stability was better, and the qualified sessions of the ointment were two years under room temperature. The studies opened up the beginning to treat clinical mastitis of dairy cow with percutaneous preparation. |
