Ougan (Citrus suavissima) is a cultivar of Rutaceae that is indigenous to southern part of Zhejiang, which is of value for health protection. Ougan is regarded to be nutritious as oryx in the traditional festival of Duanwu.This study aims to, through investigation and experiment, investigate, collect and systematically evaluate Ougan germplasm resources. All of these attempts led to selection of a superior bud mutation—seedless Ougan:1. Observations and genetic identification for consecutive showed that its desirable trails can inherit stably. The fruits are invertedly round and seedless with a shape index of 1.00 and have an average weight around 150 g. Fruit has no seed and the pulp is orange, which is delicious, juicy, fibreless. The fruits are of good flavor, superior in eating quality. The fruits are mature in middle to late November, which can be stored till May of next year without losing the flavor normal at ambient environment.2. RAPD markers were used to differentiate Ougan and seedless Ougan, which showed that primer S304 was powerful for distinguishing them from each other. The results implied seedless Ougan and Ougan were different at the level of DNA.3. Compared with Ougan, pollens of seedless Ougan could not germinate at all or pollens that emanated naturally were scarcely observed, which might be possibly the main reason causing seedlessness of the mutant.4. Shoot tip micrografting was employed to eliminate HLB in Ougan. PCR analysis showed the virus had been successfully removed.5. A serial of technologies on standard production of Ougan were established, inclusive of setting up orchard, planting, manipulating fertilizer and water, managing flower and fruit, preventing and controlling diseases and insects.
