Former analyze to the genome of rice mutant which astronautic mutagenesis confirmed that the space environment can cause the genomic polymorphisms of rice change,in order to further study the Characteristics and rules of Genomic Changes of Rice on Board Spaceship at the First Generation,we choose the seeds of BeiJingHong and LianJing-1 boarded on Shenzhou-6,in April 2006 we cultivated them in the base of Dalian institute of special grops.At the same time,rice seeds without boarding on Shenzhou-6 were planted as ground controls.During the maturity period,we selected the fresh leaves of rice as our experimental materials.This research is designed to analyze the characteristics of genomic changes of BeiJingHong and LianJing-1 at the first generation after space flight aboard Shenzhou-6 spacecraft by AFLP molecular marker.The AFLP analysis of eight ground controls without carrying on Shenzhou-6 found the genomic polymorphism were 0~0.6%and 0~0.5%between the two strains of BeiJingHong and LianJing-1 by using four pairs of primers.By using 16 pairs of AFLP selective primers,749 and 673 bands were detected in twenty plants of BeiJingHong and LianJing-1 at the first generation after aboard,respectively.About 50 bands were amplified by per pair primers.The individual differences between them were 7~49 and 8~30 bands,respectively,and the genomic DNA polymorphisms of individual plant were 0.93%~6.54%and 1.19%~4.46%for rice of the two varieties boarded on spaceship,they were all higher than its ground controls.The experimental results showed that the space environment can induce large-scale changes for rice at the first generation after aboard.The comparative analysis of experimental results discovered that the genomic DNA polymorphisms of LianJing-1 most higher than that of BeiJingHong,which reflected the mutagenic efficiency of space environment is related to the genotypes of tested materials,and the materials with different genotypes in response to the spatial environmental condition are different.In addition,the number of amplified bands with different primer combinations was varying greatly,of which the primer pair E-CC/M-TG amplified the least bands,and the bands that the primer combination E-AG/M-AT amplified were the most.The number of varied bands amplified by 16 primer combinations in BeiJingHong and LianJing-1 were also not the same,E-CA/M-AG and E-CC/M-AG have a higher efficiency to amplified different DNA bands in the two strains,but primer combinations E-AG/M-AT and E-AG/M-AA have a lower efficiency.Maybe the distribution of identified sites by different primer pairs in the genome is different,and that cause to a different number of amplified bands.Taken together,our research enriched the people' understanding on the characteristics and the laws of space environment induced the hereditary and mutation of organism.
