Ligustrum lucidum Ait., as an evergreen large shrub or tree, belongs to the Oleaceae family Ligustrum genus. It has less disease and strong resistance to sulfur dioxide, hydrogen fluoride and other harmful gases, and is one of the best green trees in the air pollution areaes. It is also a rare evergreen broad-leaved species in northern China. The present experiment studied systematically in vitro culture Ligustrum lucidum Ait. involved in a variety of factors, including the disinfection of explants; early subculture generation and screening of the basic medium, as well as hormone concentration of the best training ratio; optimal rooting medium and the hormone concentration, and the ratio of the types of options; callus induction, such as plant regeneration. The main results were as follows:1. The best picking time for explants is in April. The explants disinfectant effects are the best by using sprouting shoots and current-year shoots as explants. The best explant disinfection method is 0.1% HgCl2 with 5 min disinfectant soaking, and sterile water rinsing over 5 times.2. Explants of sprouting shoots and current-year shoots have the best results when in the initiative culture. The optimal culture medium in the beginning culture phase is MS medium. Sprouting shoots medium at the very beginning of MS +6-BA 0.5 mg / L + NAA 0.1 mg / L on the initiative has the highest sprouting rate of 80.28%; current-year shoots at the very beginning of the medium MS +6-BA 1.0 mg / L + NAA 0.05 mg / L on the initiative has the highest sprouting rate of 73.45%.3. MS is more suitable for the proliferation of cultured Ligustrum lucidum Ait. subculture than WPM and B5 is. When macro-elements in MS medium are applied for the whole amount of usage, the proliferation of Ligustrum lucidum Ait. has the highest coefficient. The germination of sprouting shoots in MS +6-BA 4.0 mg / L + NAA 0.1 mg / L achieves a coefficient of proliferation as high as 4.42.4. Macro-elements affect significantly on the rooting rate, and when they are 1/2 of MS, the average rooting rate of Ligustrum lucidum Ait. reaches its highest of 57.45%, and the average root number has reached 3.15. Formula for the optimal rooting is 1/2 MS + NAA 0.3 mg / L. The average rooting rate and the average root number are significantly higher in in vitro plants of sprouting shoots than those of current-year shoots. The average rooting rate reaches the highest of 61.34%, and the average root number is 3.58. 5. When 6-BA concentration is 3.0 mg / L and NAA concentration 0.1 mg / L, the highest rate of callus induction obtains and at the same time by using this callus induction the differentiation of adventitious buds is the highest of 9.1%.
