Distylium chinense Diels belonging to country of the second rare and endangered plants,evergreen shrub,is a very high ornamental value of bonsai,mainly in the Yangtze River Basin.In recent years because of the person is digging Distylium chinense Diels with unbridled and indiscretionary,and also river closure of the Three Gorges Project, The both reasons reduce the Distylium chinense Diels quantity.At present,there was no Distylium chinense Diels to callus induction,the paper mainly uses the leaf to callus induction.Callus growth of the optimum temperature is 20-24℃.Generally 9 days after inoculation,there will be the emergence of callus.Add none hormones in the MS medium did not induce callus.Founding that a single hormone-induced effects are relatively better on1/2MS+6-BA0.5mg/l,1/2MS+2,4-D0.01.Combination of hormone-induced effects are better than most of the role of a single hormone,in particular,to1/2MS+6-BA0.5+NAA0.5,1/2MS+6-BA0.5+2,4-D0.4.1/2MS+NAA0.1+TD Z0.2.1/2MS+2,4-D0.25+KT0.05 effect is better,the induction rate is also high,the majority can reach 100%.Callus growth in good condition,mostly green,compact, vigorous,but generally three days later at the growth secrete secondary metabolites,so at a time when is vaccinated,,pre-processed using PVP.Growth after three weeks callus will be browning,callus life expectancy is not very long,it is easy to die.These should be the reason of the Distylium chinense Diels callus that cannot be differentiation.Taking Distylium chinense Diels leaves,9 days,11 days,16 days,17 days,18 days, 20 days,24 days,26 days,28 days,30 days of callus growth and callus completely,with FAA fixed.Using paraffin section method,micro-camera at the photo,by comparison, are found in leaf palisade tissue and spongy tissue induced callus,the majority are spongy tissue.But none of them were found to participate in vascular formation of callus.
