Stropharia rugoso-annulata (King Stropharia or Winecap) which belongs to Basidiomycota, Hymenomycetes, Agaricales, Strophariaceae, Stropharia, is a gorgeous, nutritional and functional mushroom with a pleasant refreshing taste of delicate fragrance, crisp and slightly sweet. Its fruitbodies contain high protein levels with all the essential amino acids and other elements, such as trace elements, vitamins and polysaccharides which have efficacy on preventing coronary heart diseases, helping to digesting and weakening mentally exhaustion. The inhibitory rates of polysaccharides of Stropharia rugoso-annulata to S-180 tumour in mice and Ai ascites cancer achieve 70 %.Selenium (Se) is an essential bio-trace element for human, animals and some microorganisms. In human and animal bodies it acts as a component of glutathione peroxidase (GSH—Px) to break chain reaction of lipid peroxidation by catalyzing peroxide decomposition and scavenging some organic hydroperoxides, such as nucleic hydroperoxide and thimine hydroperoxide, et al., so as to maintain the integrity of structure and function of biomembrane.This paper studied the optimization of extraction technology of Se-polysaccharide produced by Stropharia rugoso-annulata in submerged culture, and analyzed its anti-oxidantal capability. The results are as follows:According to optimization experiments designed by RSM, the optimal conditions for extraction of Se-polysaccharide are: processing time 16.33min, temperature 63.1℃and pH 7.65.We got two pure components from the Se-polysaccharide named SeGL-1,SeGL-2 by the Sephadex G-100 method. The antioxidation effects on Glutathione peroxidsade (GSH—Px), Superoxide dismutase (SOD) and Malondialdehyde (MDA) of polysaccharide and Se-polysaccharide of the blood in intragastric rats were studied. The results showed that GSH-Px and SOD activities of the blood in the experimental mice with original polysaccharide and Se-polysaccharide of Stropharia rugoso-annulata were increased, and concentration of MDA was decreased sharply. The antioxidation effect of Se-polysaccharide of Stropharia rugoso-annulata was better than that of original polysaccharide.
