Follicle-stimulating hormone (FSH) is a pituitary glycoprotein gonadotropin that is composed of two subunits, the commonα-subunit and the specificβ-subunit, which determines the binding to specific receptors and induction of biological response, and it plays a key role in hypothalamic-pituitary-gonad axis.The purpose of this study was to detect single nucleotide polymorphisms (SNPs) within FSHβgene, to analyze the genetic structure of two goat breeds, and to investigate the effects of the gene on litter size of Huanghuai goats. Based on the sheep DNA sequences in GenBank, five primer pairs for FSHβgene, respectively, were designed to clone the partial sequence of FSHβgene. Detecting mutation location of exon 2 of FSHβgene in Huanghuai goat and Bore goat by PCR-SSCP, and the associations of the gene polymorphism with the litter size of Huanghuai goat does were also explored. The results are as follows:1. The PCR products of one pairs of primer among those of 5 pair primers were polymorphic. And the polymorphic site was on exon 2 of FSHβgene.2. There are 8 bases mutated on exon 2 of FSHβgene between Huanghuai goat and sheep. Homological analysis suggested that the homology of Huanghuai goat partial sequence with sheep partial sequence were 99%, respectively.3. The results indicated that three genotypes (AA, AB and AC) were detected in Huanghuai goats. Sequencing revealed C→T mutation at 47bp of exon 2 of FSHβgene and C→T mutation at 147bp of exon 2 of FSHβgene in the AB genotype in comparison to the AA genotype and C→T mutation at 147bp of exon 2 of FSHβgene and G→A mutation at 156bp of exon 2 of FSHβgene in the AC genotype in comparison to the AA genotype.4. Genetic analysis suggested that, three allelics were detected in Huanghuai goat and the allelic frequencies were 0.9257, 0.0608 and 0.0135, respectively, three genotype (AA,AB and AC) frequencies were 0.8514, 0.1216 and 0.0270, respectively.χ2 fitness test showed that the distribution of different genotype with primer 5 in Huanghuai goats was in Hardy-Weinberg equilibrium. In Bore goat, the allelic frequencies were 0.9666, 0.0444, respectively; Genotype (AA and AB) frequencies were 0.9333 and 0.0667, respectively.χ2 fitness test showed that the distribution of different genotype with primer 5 in Huanghuai goats was in Hardy-Weinberg equilibrium,too.5. In the Huanghuai goat population, least square analysis showed that the does with genotype AA had 0.37 (P>0.05) or 0.76 (P<0.05) kids more than those with genotype AB or AC, respectively; the difference between genotype AB and genotype BB was not significantly.
