This experiment use Rhodiola male plant anther as explants, study of effect of hormone on the induction and differentiation of anther callus;at the same time study of the effect of different medium and culture conditions on growth of anther callus. The results show that:1. The most suitable low temperature for Rhodiola anther culture is24hours; anther callus induction effect of NAA was better than2,4-D; the concentration of anxin NAA between0.5mg/L-5.0mg/L, the lower the concentration of NAA is more beneficial to the induction of anther callus; the medium that containing cytokinin6-BA is beneficial to the induction of anther callus.2. Different concentrations of anxin NAA have little influence on the growth of Rhodiola anther callus, in the different species of anxin2,4-D and NAA, the callus growth of2,4-D is higher than NAA, but the callus color of NAA is more green than2,4-D.3.The callus growth of higher concentrations (0.5and2.0mg/L) of cytokinin6-BA is higher than lower concentrations (0.1mg/L), anxin and cytokinin concentrations improved, the growth of callus imptoved obviously; the same concentration of different cytokinin, the medium containing6-BA is more suitable for the growth of anther callus than the medium containing KT.4. The effect of MS and NB two kinds of basic medium on the growth of Rhodiola anther callus is not significant, but the combination of every kind basic medium and specific hormone combination showed different effects, the callus growth of MS+6-BA+NAA is highest, the callus growth of NB+NAA+KT is lowest, and it exhibits a significant difference.5.In the setting five kinds of temperature gradient of this experiment, the growth of temperature(16-25℃)、20℃and25℃have the certain difference,but there was no significant difference.6. In the setting four kinds of illumination time and two kinds of hormone concentration of this experiment,16h is most beneficial to growth of Rhodiola anther callus;In the setting five kinds of light intensity of this experiment,10001ux-20001ux is more beneficial to growth of Rhodiola anther callus.7.In the setting four kinds of PH gradient of this experiment,4.5is not most beneficial to growth of Rhodiola anther callus,5.0-5.8is more beneficial to growth of Rhodiola anther callus;In the setting four kinds of sucrose concentration of this experiment,30g/L is most beneficial to growth of Rhodiola anther callus.8.5kinds of anther callus differentiation materials in the experiment, callus of MS+2. Omg/LNAA+3.0mg/L6-BA is more suitable for differentiation test; the most suitable differentiation medium in the8kinds of medium is MS+1. Omg/LNAA+2.0mg/L6-BA;in four different concentrations of NAA, higher and lower NAA concentrations both goes against anther callus differentiation; in three different concentrations of6-BA, anther callus differentiation rate increased with concentration increasing.
