Take the leaves of different sexual Rhodiola sachlinensis A.Bor as the explants to research the induction,growth and differentiation of the callus and make the careful analysis on the conditions of hormone,culture and explants,the results indicated that:(1)The medium mixed with NAA(1.5mg/L,3mg/L)and 6-BA is good for induction of female leaf callus,3.0mg/LNAA and 3.0mg/L6-BA is the best.The medium mixed with NAA(1mg/L,1.5mg/L)and 6-BA is good for induction of female leaf callus,3.0mg/LNAA and 3.0mg/L6-BA is the best.(2)According as the different select time we kwon that select female leaves at forepart,but select male leaves at metaphase;According as the different florescence,select female leaves at the time when no buds appear or buds start to appear and select male leaves at the time when no bud appear or flower open barely.Do like this,we can get the good induce effect.(3)The medium mixed with low consistency NAA and high consistency 6-BA is good for growth of male leaf callus.High consistency NAA is for the dry weight increase of male leaf callus.(4)The optimum temperature for callus growth is normal temperature 24℃.Illuminated for 12h is favorable for the male leaf callus growth.Additionally,the higher illumination intensity is advantageous to the leaf callus growth.The tightness of leaf callus increased under the low temperature,low illumination intensity and short illumination length.(5)The normal temperature 24℃,low illumination intensity(1000Lx) and Illuminated for 10h/d or 14h/d is advantageous to the male leaf callus differentiation.The medium mixed with low consistency NAA(0.1mg/L) is good for differentiation of male leaf callus.
